Abstract:
Objective: To investigate the effects of calpain inhibitor Calpeptin on the migration of MDA-MB-231 breast cancer cells for providing an idea in treating the metastatic breast cancer.
Methods: The effects of different concentrations of Calpeptin on the proliferation of MDA-MB-231 cells were determined using MTT assay.The effects of different concentrations of Calpeptin on the migration of MDA-MB-231 cells were detected using the wound healing assay and transwell migration assay.The effects of different concentrations of Calpeptin on the expression of matrix metalloproteinase-2(MMP-2) in MDA-MB-231 cells were detected using Western blot.
Results: Calpeptin could suppress the proliferation of MDA-MB-231 breast cancer cells,the inhibiting effects of which increased with the increasing of the concentrations of Calpeptin(
P<0.01).The migration ability of MDA-MB-231 cells decreased significantly after 24 h of treatment with Calpeptin(
P<0.01).The result of Western blot showed that Calpeptin could down-regulate the expression of MMP-2 in MDA-MB-231 cells.
Conclusions: Calpeptin can inhibit the migration of MDA-MB-231 cells,the mechanism of which may involve in the down-regulation of MMP-2.
