过氧化氢对人脐静脉内皮细胞白细胞介素-37表达的影响

    Effect of hydrogen peroxide on the expression of in terleukin-3 in hurnan umbilical vein endothelial cells

    • 摘要: 目的:研究过氧化氢人脐静脉内皮细胞(H2O2)对白细胞介素(IL)-37表达的影响。方法:用实时荧光定量PCR法和Western blot法检测经系列浓度H2O2作用不同时间的人脐静脉内皮细胞中IL-37 mRNA和蛋白水平的变化。结果:实时荧光定量PCR结果显示:不同浓度H2O2作用人脐静脉内皮细胞后,IL-37 mRNA水平升高,并呈浓度依赖(P<0.01)。Western blot结果表明,H2O2作用人脐静脉内皮细胞20 h后,IL-37蛋白水平较对照组显著升高(P<0.01)。结论:H2O2能够上调人脐静脉血管内皮细胞IL-37的表达,为进一步研究IL-37在细胞氧化应激状态下的作用奠定了一定基础。

       

      Abstract: Objective: To investigate the effects of hydrogen peroxide(H2O2) on the expression of interleukin-37(IL-37) in EA.hy926 cells.Methods: The expressions of IL-37 mRNA and protein in different time-points of human umbilical vein endothelial cell treated with different concentration of H2O2 were detected by qRT-PCR and Western blot.Results: The results of qPCR showed that the levels of IL-37 mRNA in human umbilical vein endothelial cell treated with different concentration of H2O2 significantly increased,which was the concentration-dependent(P<0.01).The results of Western blot showed that the level of IL-37 protein in human umbilical vein endothelial cell treated with H2O2 for 20 h was significantly higher than that in control group(P<0.01).Conclusions: The expression of IL-37 in human umbilical vein endothelial cell can be up-regulated by H2O2,which may be helpful to study the role of IL-37 in cellular oxidative stress.
