    姜黄素联合紫杉醇对肺癌细胞PC9凋亡的影响

    The effect of curcumincombinedwithpaclitaxelonlungcancercell PC9

    • 摘要: 目的:研究姜黄素联合紫杉醇对人肺癌细胞PC9凋亡的影响。方法:将不同浓度梯度姜黄素和紫杉醇单用(单药组)和联合(联合组)作用于PC9细胞,MTT法测定其对PC9的增殖抑制作用,流式细胞术检测单药和双药联合对细胞周期分布和凋亡的影响,Western blot法检测凋亡相关蛋白表达的变化。结果:两药均以时间和剂量依赖方式抑制PC9细胞的增殖,联合组抑制作用大于单药组(P<0.01)。两药均能诱导PC9细胞发生凋亡和G0/G1期阻滞,联合组细胞凋亡和G0/G1期阻滞作用显著增强(P<0.01)。Western blot法检测显示,联合组Caspase-3蛋白、Bcl-2、Bax的改变显著大于单药组(P<0.01)。结论:姜黄素、紫杉醇可抑制人肺癌PC9细胞增殖,诱导其凋亡,阻滞细胞G0/G1期,伴随Caspase-3活性上调,且两者联合应用可显著促进细胞凋亡。

       

      Abstract: Objective: To study the apoptosis-inducing effects of curcumin(CUR) combined with paclitaxel(PA) on lung cancer cell line PC9.Methods: The PC9 cells were treated with different concentrations of single drug or two drugs combination.The anti-proliferative effect of drug was measured using MTT.The effects of single drug and two drugs combination on the cell cycle distribution and apoptosis were measured by flow cytometry.The expressions of apoptosis-related protein were detected by western blotting.Results: Two drugs could inhibit the prolification of PC9 cells with time-and concentration-dependent manner,and the profilication inhibiting rate of curcumin combined with paclitaxel was higher than that of CUR or PA.The apoptosis and G0/G1 phase arrest of PC9 cells treated with drug combination significantly enhanced(P<0.01).The protein expression changes of Caspase-3,Bcl-2 and Bax in PC9 cells teated with drug combination were significantly more than that with single drug(P<0.01).Conclusions: The CUR and PA can inhibit the proliferation of human lung cancer cell PC9,induce apoptosis,block cell G0/G1 phase and inprove the Caspase-3 activity.Two drugs combination can significnatly promote the PC9 cells apoptosis.
