    Expression of circ_100290 in colorectal cancer and its clinical significance

      目的探讨环状RNA circ_100290在结直肠癌病人中的表达,及其与临床病理特征及预后的关系。
      方法实时荧光定量聚合酶链反应(qRT-PCR)法检测结直肠癌及其癌旁正常组织中环状RNA circ_100290的表达量。分析环状RNA circ_100290的表达差异与临床资料及病例特征的关系。生存曲线分析环状RNA circ_100290的表达差异与临床预后的相关性。Cox比例风险回归模型进行单变量和多变量回归分析。
      结果环状RNA circ_100290在结直肠癌组织中的相对表达水平高于癌旁肠正常组织(P < 0.01)。环状RNA circ_100290高表达与与淋巴结转移、远处转移、TNM分期有关(P < .05~P < 0.01)。生存曲线分析显示,与人环状RNA circ_100290表达低的病人相比,高表达的病人总生存的平均时间更短(P < 0.05);单变量与多变量Cox比例风险回归模型分析均显示,环状RNA circ_100290表达水平、淋巴结转移、远处转移和TNM分期与结肠癌病人的总生存率均有关系(P < 0.05~P < 0.01)。
      结论环状RNA circ_100290高表达与结直肠癌转移和预后相关。因此,环状RNA circ_100290可以作为预测结肠癌病人预后的潜在生物标志物。

       

      Abstract:
      ObjectiveTo investigate the expression level of circular RNA circ_100290 in colorectal cancer patients, and its correlation with clinicopathological features of colorectal cancer and prognosis.
      MethodsThe expression levels of circ_100290 in colorectal cancer tissue and normal tissue adjacent to cancer were detected using qRT-PCR.The relationship between circ_100290 expression level, and clinical data and case characteristics were analyzed.The correlation between circ_100290 expression level and clinical prognosis was analyzed using survival curve analysis.The Cox proportional hazard regression model was used for univariate and multivariate regression analysis.
      ResultsThe expression level of circ_100290 in colorectal cancer tissue was higher than that in normal tissue(P < 0.01), and the high expression of circ_100290 was related to the lymph node metastasis, distant metastasis and TNM stage(P < 0.05 to P < 0.01).The results of Kaplan-Meier survival analysis showed that the total mean survival time in patients with high circ_100290 expression was shorter than that in patients with low circ_100290 expression(P < 0.05).The results of univariate Cox proportional hazards regression model analysis indicated that the expression level of circ_100290, lymph node metastasis, distant metastasis and TNM stage were correlated with the overall survival rate of colorectal cancer patients(P < 0.05 to P < 0.01).
      ConclusionsThe high expression of circ_100290 is related to the colorectal cancer metastasis and prognosis.The circ_100290 may serve as a promising biomarker in predicting the prognosis of colorectal cancer.
