Abstract:
ObjectiveTo investigate the correlation of the polymorphism of fibrinogen(Fg)Bβ-249C/T gene with chronic obstructive pulmonary disease(COPD) and smoking.
MethodsEighty patients with acute exacerbation chronic obstructive pulmonary disease(AECOPD) and 100 healthy subjects were divided into the case group and control group, respectively.The level of Fg, and levels of IL-8 and TNF-α was detected using the enzyme-linked immunosorbent assay(ELISA) and radioimmunoassay, respectively.The polymorphism of FgBβ-249C/T gene was analyzed using polymerase chain reaction restriction fragment length polymorphism analysis technique, and the mutation rate of gene was calculated.
ResultsThere was genetic polymorphism in FgBβ-249C/T locus.The levels of Fg, IL-8, TNF-α, gene mutation ratio of FgBβ-249C/T locus and type CT+TT Fg level in smokers were significantly higher than those in nonsmokers in two groups(P < 0.01), and the levels of Fg, IL-8, TNF-α, gene mutation ratio of FgBβ-249C/T locus and type CT+TT Fg level in smokers of case group were significantly higher than those in smokers of control groups(P < 0.01).
ConclusionsSmoking is associated with the FgBβ-249C/T gene polymorphism and COPD.The selective polymorphism detection of the selective FgBβ-249C/T gene locus may help to screen COPD.
