胃癌组织中RASGRF1基因甲基化及蛋白表达与其临床意义

    Methylation and protein expression of RASGRF1 gene in gastric cancer and its clinical significance

      目的探讨胃癌组织中Ras蛋白特异性鸟苷酸释放因子1(RASGRF1)基因甲基化及蛋白表达以及临床意义。
      方法收集80例胃癌病人病历资料,采用免疫组织化学法检测80例胃癌组织和相应癌旁组织中RASGRF1蛋白表达情况,采用甲基化特异性PCR检测RASGRF1基因甲基化状态,分析胃癌组织中RASGRF1基因甲基化及蛋白表达的相关性,并观察胃癌组织中RASGRF1基因甲基化状态及蛋白表达情况与病人临床病理参数的关系。
      结果胃癌组织和相应癌旁组织中RASGRF1蛋白表达与基因甲基化表达情况差异均有统计学意义(P < 0.05);Spearman相关分析结果显示,胃癌组织中RASGRF1蛋白表达与基因甲基化呈负相关关系(P < 0.01);胃癌组织中RASGRF1蛋白表达与性别、年龄、肿瘤直径、组织学分化程度、发生远处转移无关(P>0.05),与肿瘤分期有关(P < 0.05);胃癌组织中RASGRF1基因甲基化与性别、年龄、肿瘤直径、组织学分化程度无关(P>0.05),与肿瘤分期、发生远处转移有关(P < 0.05和P < 0.01)。
      结论胃癌组织中RASGRF1基因甲基化和蛋白表达二者呈负相关关系,RASGRF1基因甲基化及蛋白表达与肿瘤分期、发生远处转移有一定关系。

       

      Abstract:
      ObjectiveTo investigate the methylation and protein expression of Ras specific guanylate releasing factor 1(RASGRF1) in gastric cancer and its clinical significance.
      MethodsThe clinical data of 80 patients with gastric cancer were collected.The expression levels of RASGRF1 protein in 80 gastric cancer tissues and corresponding adjacent tissues were detected using immunohistochemistry.The methylation status of RASGRF1 gene was detected by methylation specific PCR.Spearman correlation was used to analyze the correlation of methylation with protein expression levels of RASGRF1 in gastric cancer tissues.The relationship between the methylation status and protein expression levels were observed.
      ResultsThe RASGRF1 protein expression and gene methylation expression were different between gastric cancer tissues and adjacent tissues(P < 0.05).The results of Spearman correlation analysis showed that the RASGRF1 protein expression was negatively correlated with the gene methylation in gastric cancer tissues(P < 0.01).The RASGRF1 protein expression in gastric cancer tissues was not related to the gender, age, tumor diameter, histological differentiation degree and distant metastasis(P>0.05), but was related to tumor stage(P < 0.05).The RASGRF1 gene methylation in gastric cancer tissues was not related to the gender, age, tumor diameter and histological differentiation(P>0.05), but was related to the tumor stage and distant metastasis(P < 0.05 and P < 0.01).
      ConclusionsThere is a negative correlation between RASGRF1 gene methylation and protein expression in gastric cancer tissues.The RASGRF1 gene methylation and protein expression have a certain correlation with tumor stage and distant metastasis.
