    积雪草苷通过Vimentin/NLRP3通路诱导鼻咽癌细胞凋亡研究

    Asiaticoside induces apoptosis in nasopharyngeal carcinoma cells through Vimentin/NLRP3 pathway

      目的研究积雪草苷对人鼻咽癌CNE-2细胞生长、凋亡及凋亡通路的影响。
      方法体外培养CNE-2细胞,将不同浓度的积雪草苷作用于CNE-2细胞,MTT和CCK-8法检测积雪草苷对细胞生长的影响;流式细胞术检测对细胞凋亡的影响;Western blotting检测Vimentin、NLRP3、Bcl-2和Caspase-3表达;ELISA法检测TNF-α和IL-1β含量。
      结果MTT和CCK-8结果显示,积雪草苷可浓度依赖性地抑制CNE-2细胞的生长;流式细胞仪检测结果显示,积雪草苷可浓度依赖性地促进CNE-2细胞凋亡;Western blotting结果显示,积雪草苷促进Caspase-3蛋白的表达,抑制Vimentin、NLRP3和Bcl-2的表达;ELISA法结果显示,积雪草苷可浓度依赖性地降低TNF-α和IL-1β含量。
      结论积雪草苷可显著促进人鼻咽癌细胞株CNE-2细胞凋亡,其机制可能与下调Vimentin,NLRP3蛋白的表达来实现的。

       

      Abstract:
      ObjectiveTo investigate the effects of asiaticoside on the growth, apoptosis, apoptosis pathway in human nasopharyngeal carcinoma cell line CNE-2.
      MethodsThe CNE-2 cells were cultured in vitro, and treated with different concentrations of Asiaticoside.The proliferation of cells were detected using MTT and CCK-8 assay, and the apoptosis rate of cells was detected using flow cytometry.The expression levels of Vimentin, NLRP3, Bcl-2 and Caspase-3 protein were determined using Western blotting, and the serum levels of TNF-α and IL-1β were determined using ELISA.
      ResultsThe results of MTT and CCK-8 assay showed that asiaticoside could inhibit the proliferation of CNE-2 cells ina concentration-dependent manner, the results of flow cytometry showed that asiaticoside could promote the apoptosis of CNE-2 cells in a concentration-dependent manner, the results of Western blotting showed that asiaticoside promoted the expression of Caspase-3, and inhibited the expression levels of Vimentin, NLRP3 and Bcl-2, and the results of ELISA showed that the contents of TNF-α and IL-1β could be decreased in a concentration-dependent manner.
      ConclusionsAsiaticoside can significantly promote the apoptosis of human nasopharyngeal carcinoma cell line CNE-2, and the mechanism may be realized by down-regulating the expression of Vimentin/NLRP3 protein.
