肿瘤浸润淋巴细胞在Luminal型乳腺癌中的浸润特征及临床意义

    The infiltration characteristics and clinical significance of tumor infiltrating lymphocytes in luminal breast cancer

      目的 探索肿瘤浸润淋巴细胞在管腔型乳腺癌中的浸润表达及临床意义。
      方法 分析癌症基因组图谱(TCGA)中乳腺癌以及管腔型乳腺癌的原始表达数据,对管腔型乳腺癌预后相关基因进行通路富集分析以及预后分析,筛选管腔型乳腺癌的预后保护因子。
      结果 通过对TCGA数据库中乳腺癌以及管腔型乳腺癌的原始表达数据在分析,共筛选出上调基因1 003个,下调基因1 679个。进一步对管腔型乳腺癌复发组病人的预后保护相关基因的GO分析结果提示,淋巴细胞激活、T细胞受体相关通路被抑制,KEGG通路富集分析结果提示T细胞分化、抗原呈递和T细胞受体信号通路同样被明显抑制。在上述通路选择的基础上,进一步筛选出TUNAR基因是与MHC-Ⅰ类分子呈正相关关系,且与PD-L1呈负相关关系的保护性预后因子。基因相关性分析及预后相关性分析提示,TUNAR通过影响淋巴免疫相关通路在管腔型乳腺癌中发挥预后保护因子作用。
      结论 TUNAR在管腔型乳腺癌病人组织样本中低表达,且可能通过影响淋巴免疫相关通路,在管腔型乳腺癌病人预后中发挥保护作用。

       

      Abstract:
      Objective To explore the expression and clinical significance of tumor infiltrating lymphocytes in luminal breast cancer.
      Methods The original expression data of breast cancer and luminal breast cancer in the Cancer Genome Atlas(TCGA) were analyzed, and the pathway enrichment analysis and prognostic analysis were performed to screen prognostic protective factors of luminal breast cancer.
      Results By analyzing the original expression data of breast cancer and luminal breast cancer in TCGA database, 1 003 up-regulation genes and 1 679 down-regulation genes were screened out.The results of further GO analysis of prognostic protective genes in luminal breast cancer recurrence group indicated that the lymphocytes activation and T cell receptor-related pathways were inhibited, while the resulst of KEGG pathway enrichment analysis indicated that the T cell differentiation, antigen presentation and T cell receptor signaling pathways were also significantly inhibited.On the basis of the above pathway selection, TUNAR gene was further screened as a protective prognostic factor that was positively correlated with MHC-Ⅰ molecules and negatively correlated with PD-L1.The gene correlation analysis and prognostic correlation analysis showed that TUNAR might play a role as a prognostic protective factor in luminal breast cancer by affecting lymphoimmune-related pathways.
      Conclusions TUNAR is significantly low expressed in the tissue samples of patients with luminal breast cancer, and may play a protective role in the prognosis of patients with luminal breast cancer by affecting the lymphatic immune related pathways.
