慢性心理应激对裸鼠胃癌移植瘤血管生成的影响

    Effect of chronic psychological stress on angiogenesis of transplanted gastric cancer tumor in nude mice

      目的 探究慢性心理应激对裸鼠胃癌移植瘤血管生成的影响。
      方法 将人胃癌细胞株HGC-27注射于裸鼠右侧腋下构建人胃癌模型。将成功植瘤的裸鼠分为应激组及对照组,各6只。应激组建立慢性心理应激模型,对照组正常饲养。记录2组裸鼠肿瘤体积变化,测定比较血清肾上腺素、皮质酮水平,肿瘤组织内的血管内皮生长因子(VEGF)水平及微血管密度(MVD)。
      结果 与对照组比较,应激组的体质量变化、肿瘤体积、血清肾上腺素及皮质酮浓度、肿瘤组织内VEGF、MVD的水平均明显升高(P < 0.05~P < 0.01)。
      结论 慢性心理应激通过诱导肾上腺素及皮质酮的产生,促进VEGF的生成,增加裸鼠胃癌移植瘤的血管密度,加速肿瘤的生长。

       

      Abstract:
      Objective To explore the effects of chronic psychological stress on angiogenesis of transplanted gastric cancer tumor in nude mice.
      Methods Human gastric cancer cell line HGC-27 was injected into the right armpit of nude mice to establish a human gastric cancer model.The nude mice were divided into the stress group and control group(6 mice in each group).The chronic psychological stress model was established in the stress group, and the control group was raised normally.The tumor volume changes in two groups were recorded, and the serum concentrations of epinephrine and corticosterone, and levels of vascular endothelial growth factor(VEGF) and microvessel density(MVD) in tumor tissues were measured and compared.
      Results Compared with the control group, the body weight change, tumor volume, serum epinephrine and corticosterone concentrations and levels of VEGF and MVD in tumor tissues in the stress group obviously increased(P < 0.05 to P < 0.01).
      Conclusions Chronic psychological stress can induce the production of epinephrine and corticosterone, promote the production of VEGF, increase the vascular density of gastric cancer xenografts in nude mice and promote the growth of tumors.
