稽留流产病人血清、蜕膜组织中sVCAM-1、LRRFIP1表达及其与炎症、氧化应激水平的相关性

    Study on the expression levels of sVCAM-1 and LRRFIP1 in serum and decidua of patients with missed abortion and their correlation with inflammation and oxidative stress

    • 摘要: 目的: 探究稽留流产病人血清、蜕膜组织中可溶性血管细胞黏附分子-1(sVCAM-1)、富亮氨酸重复序列相互作用蛋白1(LRRFIP1)的表达及其与炎症、氧化应激水平的相关性。方法: 选取稽留流产病人72例作为研究组,另选取同期自愿终止妊娠的健康早孕妇女50例作为对照组,酶联免疫吸附法检测血清及蜕膜组织中sVCAM-1、LRRFIP1、白细胞介素(IL)-17、IL-18、IL-23以及超氧化物歧化酶(SOD)、活性氧自由基(ROS)、丙二醛(MDA)水平。Pearson法分析稽留流产病人血清及蜕膜组织中sVCAM-1、LRRFIP1水平与炎症、氧化应激指标的相关性;logistic回归分析孕妇发生稽留流产的影响因素。结果: 研究组病人血清及蜕膜组织中sVCAM-1、SOD水平明显低于对照组(P<0.01);LRRFIP1、IL-17、IL-18、IL-23、ROS、MDA水平高于对照组(P<0.01)。Pearson法分析结果显示,血清中sVCAM-1水平与IL-17、IL-18、IL-23、ROS呈负相关关系(P<0.01),与SOD呈正相关关系(P<0.01),LRRFIP1水平与IL-17、IL-18、IL-23、ROS呈正相关关系(P<0.05~P<0.01),与SOD呈负相关关系(P<0.05);蜕膜组织中sVCAM-1水平与IL-17、ROS、MDA呈负相关关系(P<0.05),与SOD呈正相关关系(P<0.05),LRRFIP1与IL-17、IL-18、IL-23、ROS呈正相关关系(P<0.05),与SOD呈负相关关系(P<0.05)。logistic回归分析结果显示,IL-17、IL-18、IL-23、ROS、MDA、LRRFIP1的升高是孕妇稽留流产的危险因素(P<0.05),SOD、sVCAM-1的升高是孕妇稽留流产的保护因素(P<0.05和P<0.01)。结论: 稽留流产病人血清及蜕膜组织中sVCAM-1呈低表达,LRRFIP1呈高表达,两者与炎症、氧化应激水平均存在一定的相关性,临床检测sVCAM-1、LRRFIP1水平对预测孕妇稽留流产的风险具有重要意义。

       

      Abstract: Objective: To investigate the expression levels of soluble vascular cell adhesion molecule-1(sVCAM-1) and leucine rich repeat interacting protein-1 (LRRFIP1) in serum and decidual of patients with missed abortion,and their correlation with inflammation and oxidative stress. Methods: A total of 72 missed abortion patients from the department of obstetrics and gynecology were selected as the study group,and 50 healthy early pregnant women who voluntarily terminated pregnancy in the same period were selected as the control group.The levels of sVCAM-1,LRRFIP1,interleukin(IL)-17,IL-18,IL-23,superoxide dismutase(SOD),reactive oxygen species(ROS) and malondialdehyde(MDA) in serum and decidua were detected by enzyme-linked immunosorbent assay(ELISA).The correlation between the levels of sVCAM-1 and LRRFIP1 in serum and decidua of missed abortion patients,and indexes of inflammation and oxidative stress were analyzed by Pearson method.The logistic regression was used to analyze the influencing factors of missed abortion in pregnant women. Results: The levels of sVCAM-1 and SOD in serum and decidua of the study group were greatly lower than those in control group(P<0.01),and the levels of LRRFIP1,IL-17,IL-18,IL-23,ROS and MDA in the study group were greatly higher than those in control group(P<0.05).The results of Pearson analysis showed that the serum level of sVCAM-1 was negatively correlated with IL-17,IL-18,IL-23 and ROS(P<0.01),and positively correlated with SOD(P<0.01);the serum level of LRRFIP1 was positively correlated with IL-17,IL-18,IL-23 and ROS(P<0.05 to P<0.01),and negatively correlated with SOD(P<0.05);the level of sVCAM-1 in decidual tissue was negatively correlated with IL-17,ROS and MDA(P<0.05),and positively correlated with SOD(P<0.01);the level of LRRFIP1 in decidual tissue was positively correlated with IL-17,IL-18,IL-23 and ROS(P<0.05 to P<0.01),and negatively correlated with SOD(P<0.05).The results of logistic regression analysis showed that the increase of IL-17,IL-18,IL-23,ROS,MDA and LRRFIP1 were the risk factors of missed abortion in pregnant women(P<0.05),and the increase of SOD and sVCAM-1 were the protective factors of missed abortion in pregnant women(P<0.05 and P<0.01). Conclusions: The expression levels of sVCAM-1 in serum and decidua of missed abortion patients are low,and the expression levels of LRRFIP1 are high,both are correlated with the levels of inflammation and oxidative stress.The clinical detection of sVCAM-1 and LRRFIP1 levels is of great significance in predicting the risk of missed abortion of pregnant women.
