miR-218-5p和HOXB3在口腔鳞癌中的表达关系研究

      目的 检测微小RNA-218-5p(miR-218-5p)和同源盒B3(HOXB3)在口腔鳞状细胞癌(OSCC)组织中的表达及其意义。
      方法 选择87例OSCC病人的OSCC组织标本及癌旁组织标本,检测miR-218-5p、HOXB3 mRNA表达水平及HOXB3蛋白表达水平;Pearson法分析miR-218-5p与HOXB3 mRNA的相关性;双荧光素酶报告基因实验验证miR-218-5p和HOXB3的靶向关系;Kaplan-Meier生存曲线分析miR-218-5p、HOXB3表达与OSCC病人生存率关系;Cox回归模型分析OSCC病人的预后影响因素。
      结果 OSCC组织中miR-218-5p表达水平明显低于癌旁组织(P < 0.01),HOXB3 mRNA表达水平明显高于癌旁组织(P < 0.01)。OSCC组织中HOXB3蛋白阳性表达率明显高于癌旁组织(P < 0.01)。Pearson相关分析显示,OSCC组织中miR-218-5p和HOXB3 mRNA表达呈负相关关系(r=-0.581,P < 0.05)。双荧光素酶报告基因实验证实miR-218-5p和HOXB3存在靶向关系。OSCC组织中miR-218-5p、HOXB3表达与TNM分期和淋巴结转移有关(P < 0.05~P < 0.01)。miR-218-5p高表达病人3年总生存率明显高于miR-218-5p低表达病人(P < 0.01);HOXB3阳性表达病人3年总生存率明显低于HOXB3阴性表达病人(P < 0.01)。淋巴结转移、miR-218-5p、HOXB3是OSCC病人不良预后的独立危险因素(P < 0.05~P < 0.01)。
      结论 OSCC组织中miR-218-5p低表达、HOXB3高表达,二者均与OSCC病人临床病理特征和预后有关,可能在OSCC发生、发展中发挥重要作用。

       

      Abstract:
      Objective To detect the expression of microRNA-218-5p (miR-218-5p) and homeobox B3 (HOXB3) in oral squamous cell carcinoma (OSCC) tissues and its clinical significance.
      Methods Eighty-seven OSCC tissue specimens and corresponding paracancerous tissue specimens were selected.The expression levels of miR-218-5p, HOXB3 mRNA, and HOXB3 protein were detected.Pearson method was used to analyze the relationship between miR-218-5p and HOXB3 mRNA.The targeting relationship between miR-218-5p and HOXB3 was verified through dual luciferase reporter gene experiment.The relationship between miR-218-5p, HOXB3 expression and survival rate of OSCC patients was evaluated by Kaplan-Meier survival curve.The influencing factors for prognosis in OSCC patients were analyzed with Cox regression model.
      Results The expression level of miR-218-5p in the OSCC tissues was significantly lower than that in the paracancerous tissues (P < 0.01), and the expression level of HOXB3 mRNA was significantly higher than that in the paracancerous tissues (P < 0.01).The positive expression rate of HOXB3 protein in the OSCC tissues was significantly higher than that in the paracancerous tissues (P < 0.01).Pearson correlation analysis showed that the expression of miR-218-5p mRNA was negatively correlated with the expression of HOXB3 mRNA in the OSCC tissues (r=-0.581, P < 0.05).The dual luciferase reporter gene experiment confirmed the targeting relationship between miR-218-5p and HOXB3.The expression of miR-218-5p and HOXB3 in OSCC tissues was associated with TNM staging and lymph node metastasis (P < 0.05 to P < 0.01).The 3-year overall survival rate of patients with high miR-218-5p expression was significantly higher than that of patients with low miR-218-5p expression (P < 0.01);the 3-year overall survival rate of patients with positive HOXB3 expression was significantly lower than that of patients with negative HOXB3 expression (P < 0.01).Lymph node metastasis, miR-218-5p, and HOXB3 were independent risk factors for poor prognosis in OSCC patients (P < 0.05 to P < 0.01).
      Conclusions MiR-218-5p is lowly expressed, and HOXB3 is highly expressed in OSCC tissues, both are related to the clinicopathological characteristics and prognosis of OSCC patients, and may play an important role in the occurrence and development of OSCC.
