Nrf2、HO-1、Notch1、Hes-1在糖尿病视网膜病变病人玻璃体和血清中的表达及意义

    Expression and significance of Nrf2, HO-1, Notch1, Hes-1 in vitreous and serum of patients with diabetic retinopathy

      目的 探讨核因子E2相关因子2(Nrf2)、血红素加氧酶-1(HO-1)、缺口受体1(Notch1)、发状分裂相关增强子1(Hes-1)在糖尿病视网膜病变病人玻璃体和血清中的表达及意义。
      方法 收集进行玻璃体切割手术的糖尿病视网膜病变病人84例,其中糖尿病视网膜病变病人43例作为观察组,单纯性孔源性视网膜脱离41例作为对照组。比较2组玻璃体和血清中Nrf2、HO-1、Notch1、Hes-1的表达水平;Pearson相关分析Nrf2、HO-1、Notch1、Hes-1水平与临床指标的相关性。
      结果 与对照组相比,观察组血清FBG、HbA1c、TC、TG、MDA高表达(P < 0.01),SOD低表达(P < 0.01)。观察组血清和玻璃体中Nrf2、HO-1、Notch1、Hes-1水平均明显低于对照组(P < 0.01)。相关性分析显示,Nrf2、HO-1、Notch1、Hes-1水平与FBG、HbA1c、TC、TG、MDA呈明显负相关关系(P < 0.01);与SOD水平呈明显正相关关系(P < 0.01)。
      结论 糖尿病视网膜病变病人玻璃体和血清中的Nrf2、HO-1、Notch1、Hes-1均显著低表达,其表达与病人FBG、HbA1c、TC、TG、MDA、SOD等临床指标显著相关。

       

      Abstract:
      Objective To investigate the expression and significance of nuclear factor E2 related factor 2 (Nrf2), heme oxygenase-1 (HO-1), Notch1 and hairy and enhancer of split homolog 1 (Hes-1) in vitreous and serum of patients with diabetic retinopathy.
      Methods Eighty-four patients with diabetic retinopathy treated with vitrectomy were collected. Among them, 43 patients with diabetes retinopathy were served as the observation group, and 41 patients with simple rhegmatogenous retinal detachment were served as the control group. The expression levels of Nrf2, HO-1, Notch1, Hes-1 in the vitreous and serum between two groups were compared. Pearson method was used to analyze the correlation between the levels of Nrf2, HO-1, Notch1 and Hes-1 and clinical indicators.
      Results Compared with the control group, the serum FBG, HbA1c, TC, TG and MDA in the observation group were significantly overexpressed (P < 0.01), the SOD was obviously underexpressed (P < 0.01). The levels of Nrf2, HO-1, Notch1, Hes-1 in the serum and vitreous of the observation group were obviously lower than those of control group (P < 0.01). The results of correlation analysis showed that the levels of Nrf2, HO-1, Notch1, Hes-1 were significantly negatively correlated with FBG, HbA1c, TC, TG, and MDA (P < 0.01), and positively correlated with SOD level (P < 0.01).
      Conclusions The expression of Nrf2, HO-1, Notch1, Hes-1 in vitreous and serum of patients with diabetesic retinopathy is obviously lower, and their expression is significantly correlated with clinical indicators such as FBG, HbA1c, TC, TG, MDA, SOD.
