重组人粒细胞巨噬细胞集落刺激因子凝胶对深Ⅱ度烧伤创面溶痂的影响

    The effect of recombinant human granulocyte-macrophage colony-stimulating factor on the debridement of deep partial-thickness burn

    • 摘要: 目的:验证重组人粒细胞-巨噬细胞集落刺激因子(rhGM-CSF)对深Ⅱ度烧伤创面坏死组织溶痂的作用效果,并分析其可能的机制。方法:将70只SD大鼠用热液烧伤的方法(75℃,8 s)制成背部深Ⅱ度烧伤创面,将烧伤大鼠随机分为实验组和对照组各35只。对照组创面局部外用不含rhGM-CSF的凝胶基质,实验组创面局部外用rhGM-CSF凝胶(100 μg/10 g)。分别于制创后1、3、5、7、10、14和21 d观察2组动物创面情况并摄像;计算不同时相点的溶痂率;免疫组织化学染色观察创面中性粒细胞和巨噬细胞浸润情况。结果:烧伤后第5~10天,实验组大鼠创面溶痂率均明显高于对照组(P<0.01);创面免疫组织化学检查显示烧伤第3~7天,实验组创面局部炎症细胞浸润均较对照组明显增多(P<0.01)。结论:外源性rhGM-CSF的应用可促进深Ⅱ度烧伤创面早期炎症细胞的浸润,促进创面溶痂,从而促进深Ⅱ度烧伤创面愈合。

       

      Abstract: Objective: To explore the effects of recombinant human granulocyte-macrophage colony-stimulating factor(rhGM-CSF) on the debridement of deep partial-thickness burn,and its mechanism.Methods: Seventy SD rats with deep partial-thickness burn were established using thermal-water burn(75℃,8 s),and were randomly divided into the experiment group(35 rats) and control group(35 rats).The burn wounds of control group and experiment group were treated with matrix gel and rhGM-CSF gel,respectively.The wounds and dissolving rate of eschar in two groups were analyzed at 1,3,5,7,14 and 21 days after injury.The neutrophils and macrophages infiltration of wounds were observed using immunohistological staining.Results: The dissolving rate of wound eschar in experiment group was significantly higher than that in control group from day 5 to day 10(P<0.01).The inflammatory cells infiltration of wound in experiment group was higher than that in control group from day 3 to 7 day(P<0.01).Conclusions: The rhGM-CSF can promote the inflammatory cells infiltration,debridement and healing of deep partial thickness burn wound.
