Role of ERK-1/2 in restenosis of vein grafts

摘要: 目的:研究细胞外信号调节激酶-1/2(extracellular regulated protein kinases,ERK-1/2)活化在移植静脉中的作用。方法:用45只健康成年大白兔建立自体颈外静脉翻转行颈动脉转流手术模型。随机分成正常组、VG组和VG/UO组,Western blot分析移植静脉用来做ERK-1/2活化。增殖细胞核抗原(proliferating cell nuclear antigen,PCNA)检测细胞增殖的指标。结果:VG组移植静脉28天后发现内膜增生,静脉移植动脉化导致了ERK-1/2表达的双峰(3 h,3 d)。PCNA在术后3 h开始表达,持续到28天。而应用UO126(ERK-1/2抑制剂)后,ERK-1/2活化被明显抑制,减弱了其在细胞增殖和细胞凋亡中的作用。结论:ERK-1/2的激活可能是内膜增生的关键环节,通过抑制ERK-1/2的活化可以抑制内膜增生,防止移植血管再狭窄。Abstract: Objective:To study the role of extracellular regulated protein kinases (ERK-1/2) in vein grafts.Methods:Forty-five adult rabbits were randomly divided into control group,VG group and VG/UO group.Carotid artery bypasses were performed using reversed autologous external jugular vein.A segment of each sample was analyzed with western blot.Results:Intimal hyperplasia was observed 28 days after the operation in group VG.The reversed autologous external jugular vein grafts resulted in activation of ERK-1/2 with stronger activation occurring at 3 h and at 3 d.Proliferating cell nuclear antigen (PCNA) began to express 3 h after the operation and lasted for 28 d.The activation of ERK-1/2 in group VG/UO was significant inhibited after the application of UO126,and there was no peak of activation detected.Conclusions:ERK-1/2 activation may be the key point in the process of intimal hyperplasia.Inhibition of the ERK-1/2 pathway inhibits intimal hyperplasia,and therefore,results in inhibition of the late events.
