    • 摘要: 目的: 探讨occludin蛋白的表达与人乳腺癌细胞株MCF-7、MDA-MB-231、SKBR-3恶性增殖之间的关系。方法: 四甲基偶氮唑蓝法检测细胞增殖,流式细胞术检测细胞周期,FN蛋白黏附实验检测细胞黏附力,Transwell检测细胞迁移,RT-PCR检测occludin mRNA的表达,Western blot、细胞免疫化学技术检测occludin蛋白表达。结果: MCF-7细胞的增殖速度、S期所占比例、细胞黏附以及细胞迁移均小于MDA-MB-231和SKBR-3细胞(P < 0.05~P < 0.01),而MCF-7细胞中occludin mRNA及蛋白表达水平均高于MDA-MB-231和SKBR-3细胞(P < 0.01)。结论: occludin表达与人乳腺癌细胞恶性增殖、黏附及迁移呈一定的相关性。

       

      Abstract: Objective: To study the relationship between the expression of occludin and malignancy multiplication in different human breast cancer cell lines MCF-7,MDA-MB-231 and SKBR-3. Methods: Cell proliferation was determined by MTT assay. The cell cycle was measured by flow cytometry. The adherence was detected by fibronectin protein. The migration of cells was detected by Transwell. The expression of occludin mRNA was examined by semi-quantitative RT-PCR. Western-blot and immunohistochemistry were used to detected the protein expression of occludin. Results: Human breast cancer cell lines MCF-7 cell malignant proliferation. S phese was less than the propotion of human breast cancer cell lines MDA-M B-231 and SKBR-3 (P < 0.05-P < 0.01). The expressions of occludin mRNA and protein in the human breast cancer cell line MCF-7 were higher than that of human breast cancer cell lines MDAMB-231 and SKBR-3(P < 0.01),and the migration and adherence was weaker than that of human breast cancer cell lines MDA-MB-231 and SKBR-3(P < 0.01). Conclusions: The expression of occludin is negatively related with human breast cancer cells malignant proliferation growth,adherence and migration.
