    Evaluation on performance of reticulocytes detection by flow cytometry

    • 摘要: 目的:评价流式细胞仪测定外周血中网织红细胞(reticulocyte,Ret)百分比的性能。方法:应用流式细胞仪检测外周血Ret百分比,进行准确性、重复性及稳定性测试,并同时与手工计数结果比较,进行线性评价。结果:流式细胞仪检测外周血Ret百分比日间精密度与日内精密度CV均<0.36%,稳定性好;4℃低温24h内Ret百分比计数CV<0.43%,随着样本放置时间的延长,计数结果略有上升,不影响临床诊断;与同期手工法结果呈正相关关系(P<0.01)。结论:流式细胞仪测定Ret百分比准确性高,重复性、稳定性好,速度快,灵敏度高,纯度高,能满足临床需要,可替代手工计数。

       

      Abstract: Objective: To evaluate the detection of reticulocytes by flow cytometry comprehensively.Methods: The percent of peripheral blood reticulocytes was detected by flow cytometry.The precision reproducibility and stability were observed.The results were compared with manual method and evaluated linearly.Results: Precision CV of the indexes was less than 0.36%,which had a good stability.And the percentage CV at 4℃ in 24 hours was less than 0.43%.With the prolongation of storage time of samples,the results increased slightly,but no effect on clinical diagnosis.It had a positive correlation with manual methods (P<0.01).Conclusions: Analyzing reticuloctyes by flow cytometry is a simple,fast,accurate,stable method and has a good reproducibility.It can meet clinical requirement and replace manual methods.
