Abstract:
Objective: To evaluate manual microcolumn gel immune method in detecting antigens of blood groups of ABO and RhD.
Methods: Fifty samples of fresh anticoagulant blood,50 samples of fresh non-anticoagulant blood,50 samples of blood stocked for 15 days,but within valid period and 50 samples of non-fresh blood stocked for 3-5 days(possibly contaminated blood),which were collected from different departments,diseases and genders of patients,were classified into Ⅰ,Ⅱ,Ⅲ and Ⅳ groups respectively.Blood antigens of ABO,RhD in all samples were assayed by manual microcolumn gel immune method and tube method.The response result and response intension were compared.
Results: The result by manual microcolumn gel immune method in Ⅰ,Ⅱ and Ⅲ groups was identical with that by tube method in detecting antigens of blood type of ABO and RhD(
P>0.05),but the response intension was stronger by manual microcolumn gel immune method than that by tube method.Two improper positive samples in Ⅳ group were found by manual microcolumn gel immune method and the other 48 samples had the same results by two methods(
P>0.05).
Conclusions: Manual microcolumn gel immune method can substitute tube method for detecting antigens of blood groups of ABO and RhD.
