三氧化二砷对小鼠恶性黑素瘤抑制机制的探讨

    Inhibitory mechanism on the mice melanoma by arsenic trioxide

    • 摘要: 目的:探讨三氧化二砷(As2O3)对小鼠B16细胞实体瘤的生长抑制作用及对端粒酶活性表达的影响,旨在为临床治疗黑素瘤提供理论和实验依据。方法:以不同浓度As2O3作用的鼠黑素瘤B16细胞荷瘤小鼠,用TRAP-PAGE-银染法检测药物作用后瘤体组织端粒酶活性的表达;通过测定荷瘤小鼠瘤体大小和肿瘤组织内微血管密度来反映不同剂量As2O3的抑瘤作用。结果:As2O3能抑制鼠B16细胞实体瘤的生长,破坏肿瘤组织微血管的形成(P<0.01);As2O3能明显抑制鼠B16细胞实体瘤端粒酶的活性。结论:As2O3对于B16细胞实体瘤的生长有显著的抑制作用,这可能与其抑制肿瘤组织内微血管的形成和端粒酶表达活性有关。

       

      Abstract: Objective: To investigate inhibitory effects of arsenic trioxide(As2O3) on the growth of B16 cell solid tumours in C57BL/6J mice and telomerase activity;and to provid experiment basis for treatment of malignant melanoma.Methods: Mouse melanoma B16 cells were transplanted into C57BL/6J mice that were treated with different concentrations of As2O3.The weight of tumor and percent of tumor development were examined.The telomerase activity in mice tumours was detected by TRAP-PAGE-silver staining.Results: The tumor growth was significantly inhibited by As2O3.The microvessel density in tumor tissues was obviously reduced by As2O3(P<0.01).Treated with As2O3,the telomerase activity was obviously reduced.Conclusions: As2O3 can inhibit the growth and the telomerase activiy of B16 cell solid tumours in C57BL/J mice.
