    Influence of arsenic trioxide on the expression of telomerase activity in melanoma B16 cells

    • 摘要: 目的: 研究三氧化二砷(As2O3)对黑色素瘤B16细胞端粒酶活性的作用,探讨砷及其化合物治疗黑色素瘤的作用机制,为其治疗黑色素瘤提供新的理论和实验依据。方法: 用TRAP-ELISA和TRAP-PAGE银染法检测黑色素瘤B16细胞(简称B16)端粒酶活性。结果: As2O3对黑色素瘤B16细胞端粒酶活性的抑制有明显的浓度和时间依赖关系。结论: As2O3对黑色素瘤B16细胞端粒酶活性表达的抑制作用随药物浓度的增加或作用时间的延长而增强。

       

      Abstract: Objective: To study the influence of arsenic trioxide(As2O3) on the telomerase activity in melanoma B16 cells,so as to provide new theoretical and experimental basis for the treatment of melonoma by As2O3.Methods: The telomerase activity in melonoma B16 cells was detected by TRAP-ELISA and TRAP-PAGE-silver staining.Results: As2O3 could inhibit telomerase activity in melanoma B16 cells and the rates of inhibition had both dose-dependent and time-dependent manner.Conclusions: The inhibition of B16 cells by As2O3 is enhanced with the increase of As2O3 concentration or prolongation of co-culture time.
