胃癌RASSF1A的表达与幽门螺杆菌感染关系的研究

    Expression of RASSF1A and infection of Helicobacter pylori in human primary gastric carcinomas

    • 摘要: 目的:探讨RAS相关区域家族基因1A(RASSF1A)在胃腺癌中的表达及与幽门螺杆菌(Hp)感染的关系。方法:用逆转录-聚合酶链反应(RT-PCR)方法检测48例胃癌组织及15例癌旁正常组织中RASSF1A mRNA的表达,用PCR法检测上述胃癌组织中的Hp感染情况。结果:RASSF1A在胃癌组织中转录表达缺失率为50.0%,其表达缺失率与胃癌分化程度、淋巴结转移及TNM分期有一定关系(P<0.05),与其组织学类型无明显关系(P>0.05)。胃癌组织中Hp感染率为62.5%(30/48),RASSF1A的表达缺失与Hp感染组之间无相关性(P>0.05)。结论:RASSF1A是胃癌的相关抑癌基因,RASSF1A的表达缺失与胃癌的临床分期和病理分级具有一定关系,RASSF1A的表达缺失与Hp感染之间无明显关系,它们可能是两个独立的致病因素。

       

      Abstract: Objective: To investigate the relationship between the expression of RASSF1A in human primary gastric adenocarcinomas and the infection of Helicobacter pylori(Hp).Methods: The mRNA expression of RASSF1A was determined by reverse transcription polymerase chain reaction(RT-PCR) in 48 cases of primary gastric carcinoma and 15 matched normal tissues.Hp was examined in 48 cases of gastric carcinoma by polymerase chain reaction(PCR) technique.Results: The rate of loss of RASSF1A mRNA expression in 48 primary gastric carcinomas was 52%(26/48),and positively related with histological differentiation degree(P<0.05),lymph node metastasis(P<0.05) and TNM stages(P<0.05).There was no relationship between RASSF1A expression and Hp infection(P>0.05).Conclusions: RASSF1A is a related tumor suppressor gene of gastric carcinoma,it is closely correlated with the tumor clinical pothological stage and the histological differentiation degree.There is no relationship between RASSF1A expression and Hp infection,both are probably independent etiological factors.
