阿霉素中毒大鼠肺组织中β-防御素-2的基因表达

    Expression of β-defensin-2 gene in the lung tissue of rats intoxicated by adriamycin

    • 摘要: 目的:探讨阿霉素(adriamycin,ADM)所致大鼠急性肺损伤对肺组织β-防御素-2(BD-2)基因表达的影响。方法:雄性SD大鼠40只,随机分成ADM低剂量组(10 mg/kg)、中剂量组(20 mg/kg)、高剂量组(40 mg/kg)及对照组,每组10只。模型组分别一次性腹腔注射不同剂量ADM制备急性肺损伤模型;对照组用生理盐水腹腔注射。3天后剖胸取肺,采用半定量RT-PCR技术检测肺组织BD-2 mRNA表达的变化。结果:ADM中、高剂量组大鼠BD-2基因表达均较对照组明显上调(P<0.01),低剂量组与对照组无统计学意义(P>0.05)。结论:ADM所致大鼠急性肺损伤中,肺组织BD-2的mRNA表达水平上调。

       

      Abstract: Objective: To observe the influence of acute pulmonary injury induced by adriamycin(ADM) on β-defensin-2(BD-2) gene expression in rats.Methods: Forty male Sprague-Dawley rats were randomly divided into four groups:low dose ADM group,medium dose group,high dose group and control group.The ADM groups were administered ADM of different doses(10,20 and 40 mg·kg-1·d-1).Meanwhile,normal saline was given to the control group.All the rats were killed three days later,and the lung tissues were removed by thoracotomy.The expression of BD-2 at mRNA level was determined by RT-PCR analysis.Results: The level of BD-2 gene expression in medium-dose and high-dose groups was up-regulated significantly compared with that in the control group(P<0.01);and the difference had no statistical significance between the low-dose group and the control(P>0.05).Conclusions: The level of BD-2 gene is up-regulated in the lung tissues of rats with acute pulmonary injury induced by ADM.
