弓形虫病三种病原学检查方法的比较

    Comparation of mathods for pathogenic diagnosis of toxoplasmosis

    • 摘要: 目的: 观察三种检测方法检测培养中的细胞被弓形虫感染的感染率差异,以寻找一种能够提高弓形虫病病原学诊断的敏感性方法。方法: 以弓形虫速殖子加入分孔培养的Hela细胞和THP-1细胞中,分别用吉姆萨染色、间接荧光(IFA)染色和流式细胞仪(FCM)三种方法在不同培养时间内检测宿主细胞感染弓形虫速殖子的感染率。结果: 吉姆萨染色检测平均感染率为22.0‰,IFA检测平均感染率为24.3 ‰,两者均低于FCM检测平均感染率42.0 ‰(P<0.05)。结论: 三种方法中以FCM法检测感染率为最高,I FA法和吉姆萨染色法相似。

       

      Abstract: Objective: To compare three methods of detecting Toxoplasma-gondii-infected-cells in culture and to explore a better one for pathogenic diagnosis of toxoplasmosis.Methods: The Toxoplasma tachyzoites were added in the Hela cell and THP-1 cell culture system,then the infected cells were counted at different times(3 to 9 hours) by light microscope with the Giemsa staining,by fluorescence microscope with IFA,and by flow cytometry(FCM).Results: The infection rates of Hela cells examined by Giemsa staining and by IFA were 22.0‰ and 24.3‰ respectively,and the infection rate of THP-1 cells examined by FCM was 42.0‰(P<0.05). Conclusions: The detection rate of affected THP-1 cells is the highest by FCM,while it is similar by IFA or Giemsa staining.
