ERK1/2在姜黄素对肺癌A549细胞增殖抑制中的作用

摘要: 目的: 探讨姜黄素在抑制肺癌A549细胞增殖过程中,对ERK1/2丝裂素活化蛋白激酶(MAPK)信号转导通路的影响。方法: 不同浓度的姜黄素作用于A549细胞24 h后,采用MTT法检测姜黄素对A549肺癌细胞增殖的抑制作用;应用Westernblot分别检测ERK/2、p-ERK/2蛋白及其下游相关基因基质金属蛋白酶-9(MMP-9)、组织金属蛋白酶抑制物-1(TIMP-1)的表达。结果: 姜黄素对A549细胞增殖有明显的抑制作用,且具有剂量依赖性(P<0.01)。姜黄素抑制A549细胞p-ERK蛋白的表达(P<0.05),但不影响总ERK1/2蛋白的表达;姜黄素可促进TIMP-1蛋白表达(P<0.05),但抑制MMP-9的表达(P<0.01)。结论: 姜黄素影响ERK1/2MAPK信号转导通路,调节凋亡相关蛋白表达是其抑制A549细胞细胞增殖的作用之一。Abstract: Objective: To study the role of ERK1/2 MAPK signal pathway in the proliferation-inhibitory effect of curcumin on A549 lung cancer cells. Methods: The proliferation-inhibitory effect of curcumin on A549 cells was assessed with method of MTT assay. Levels of ERK1/2,p-ERK1/2,TIMP-1 and MMP-9 protein expression were measured by Western blot. Results: Curcumin inhibited A549 cells proliferation in a dose-dependent manner. The level of p-ERK1/2、MMP-9 decreased,and the expression of TIMP-1 protein increased with the dose of curcumin but the expression of ERK1/2 protein had no change. Conclusions: Curcumin could influence the ERK1/2 MAPK signal pathway,induce the expression of apoptosis-related proteins and inhibit the proliferation of A549 cells.
