核因子-B寡聚脱氧核苷酸诱骗剂处理树突状细胞对大鼠胶原诱导性关节炎治疗作用研究

摘要: 目的:探讨核因子-B(nuclear factor-B,NF-B)寡聚脱氧核苷酸(ODN)诱骗剂处理的树突状细胞(dendritic cells,DC)对Ⅱ型胶原诱导关节炎(collagen induced arthritis,CIA)大鼠的治疗作用及机制。方法:建立Ⅱ型胶原诱导大鼠关节炎模型,NF-B诱骗剂处理SD大鼠脾脏来源的DC,在初次免疫第5天经尾静脉注射到CIA大鼠体内,并设空白对照组、CIA模型组和Decoy-DC组。42 d后观察各组关节肿胀程度、关节炎指数和病理指标。结果:经rr GM-CSF和rr IL-4刺激后诱导的各组DC表面OX62的表达在70%以上,差异无统计学意义(P0.05);Decoy-DC组与LPS-DC组的CD80阳性细胞百分率差异有统计学意义(P0.05),在Decoy-DC组的CD80荧光强度明显低于LPS-DC组(P0.01);Decoy-DC组的CD86荧光强度亦均明显低于LPS-DC组(P0.01),初次免疫第7天后,Decoy-DC组和CIA模型组的关节炎指数开始增高;从第21天后开始,Decoy-DC组的关节指数均明显低于CIA模型组(P0.01)。结论:NF-B诱骗剂处理的DC有效地减轻CIA大鼠关节炎症状及组织病理损害,对类风湿性关节炎有较好的治疗作用。Abstract: Objective:To explore the effects and mechanism of dendritic cells(DC) induced by nuclear factor-B(NF-B) oligodeoxynucleotide(ODN) in rats with type Ⅱ collagen-induced arthritis(CIA).Methods:The model rats with arthritic were established by type Ⅱ collagen inducing.DC from rat spleen treated with NF-B ODN were injected to caudal vein of CIA rats at 5 days after the initial immunization.The change of paw-swelling,the index of arthritis and pathogenic indicator of the control group,CIA model group and experimental group were observed after 42 days.Results:The OX62 expression on the DC surface of three groups were above 70%,there was no significant difference(P0.05).The CD80 postive cells of CIA model group and experimental group had significant difference(P0.01).The CD80 and CD86 average fluorescence intensity of experimental group was significantly higher than that of CIA model group(P0.01).At 7 days primary immune,the arthritis index of CIA model group and experimental group began to increase,the arthritis index of experimental group were significantly lower than that of CIA model group at 21 days(P0.01).Conclusions:The DC induced by NF-B ODN can effectively relieve the arthritis symptoms and histopathology damage of CIA rats,and have good curative effects on rheumatoid arthritis.
