Abstract:
Objective To investigate the relationship between the expression of mir-153-3p and Golgi integral membrane protein 4 (GOLIM4) in nasopharyngeal carcinoma tissues and clinicopathological characteristics of patients.
Methods A total of 102 nasopharyngeal carcinoma patients treated with surgery were selected as the observation group, and the cancer tissues of surgical patients were collected. One hundred and two patients with lymphoid tissue hyperplasia were detected using the nasopharyngeal biopsy, the chronic inflammation tissues (no nasopharyngeal carcinoma tissues) were selected as the control group. The Real-time fluorescence quantitative PCR (qRT-PCR) was used to detect the miR-153-3p level, and the immunohistochemical staining was used to detect the distribution of GOLIM4. The level of GOLIM4 in tissues was detected by Western blotting. The relationship between the expression of miR-153-3p and GOLIM4 and clinicopathological indexes was analyzed. The Kaplan-Meier method was used to analyze the relationship between the expression levels of miR-153-3p and GOLIM4 and prognosis of patients.
Results The expression level of GOLIM4 in cancer tissues was higher than that in the control group, and the expression level of mir-153-3p was lower than that in the control group, and the expression of GOLIM4 was negatively correlated with mir-153-3p (r = –0.523, P < 0.05). The expression of GOLIM4 and mir-153-3p in nasopharyngeal carcinoma tissues were not related to the age, gender, tumor diameter and pathological type, but related to the differentiation, lymph node metastasis, distant metastasis, clinical stage and T stage (P < 0.05). The results of Kaplan Meier analysis showed that the 3-year cumulative survival rate in the high GOLIM4 expression group was significantly lower than that in low expression group (P < 0.05), and the 3-year cumulative survival rate in the mir-153-3p high expression group was significantly higher than that in low expression group (P < 0.01).
Conclusions The expression of mir-153-3p is low, and the GOLIM4 is high in the cancer tissues of patients with nasopharyngeal carcinoma. The expression of mir-153-3p and GOLIM4 are closely related to the differentiation degree, lymph node metastasis, distant metastasis, clinical stage, T stage and prognosis of patients with nasopharyngeal carcinoma.
