腹腔注射卡因酸对大鼠海马细胞凋亡的影响

    Influence of intraperitoneal administration of kainite on hippocampal neurons of rats

    • 摘要: 目的: 探讨腹腔注射兴奋性氨基酸对大鼠海马神经元及Bax/bcl-2表达的影响。方法: 将大鼠随机分成两组,实验组给予腹腔注射卡因酸,对照组给予腹腔注射等体积磷酸盐缓冲液(PBS);3天后以尼氏染色海马神经元,T U NEL法检测海马神经元凋亡;Bax/bcl-2抗体标记神经元Bax/bcl-2蛋白,SABC法检测。结果: 实验组大鼠海马神经元带变狭,TUNEL染色阳性对照片可见大量阳性细胞,阴性对照片未见阳性细胞;实验组Bax为强阳性,Bcl-2为阳性;PBS生理盐水对照组Bax为弱阳性(+),Bcl-2均为强阳性(++)。结论: 腹腔注射卡因酸可以引起大鼠海马神经元凋亡,Bax表达上调,Bax/bcl-2平衡改变可能与神经元凋亡有关。

       

      Abstract: Objective: To study the influence of kainate on the hippocampal neuron of rats and the expression of Bax/bcl-2.Methods: The rats were divided into two groups random ly.The ex perimental groupw as given intraperitoneal administration of kainate, and the control groupw as given phosphate-buffered saline solution (PBS).After 3 days, the loss of neuron in the hippocampus of rats was detected by Nissl staining.The neuronic apoptosis in the hippocampus was examined with terminal deoxy nucleotidyl-transferase mediated d-U TP-biotin nick end labeling (TUN EL);related gene Bax and Bcl-2 expression were marked by SABC method.Results: Kainate could induce neuron apoptosis and loss, up-regulation of bax and down-reg ulation of bcl-2 in the hippocampus of rats by intraperitoneal administration.Conclusions: The hippocampal neuron of rats can be impaired by intraperitoneal administration of kainate.Elevated expression of Bax/bcl-2 of neuron mig ht be involved in the pathophysiologic course of apoptosis in neurons.
