    蚌埠市汉族人群尾加压素Ⅱ基因S89N变异与2型糖尿病及肥胖的相关性

    Association of urotensin Ⅱ gene S89N polymorphism with type 2 diabetes and obesity among the Han population in Bengbu area

    • 摘要: 目的:探讨蚌埠市汉族人群尾加压素Ⅱ(UⅡ)基因多态性以及与2型糖尿病(T2DM)和肥胖的相关性。方法:随机选择蚌埠市汉族人群T2DM患者105例(其中肥胖组53例,非肥胖组52例),非糖尿病(ND组)114例(其中肥胖组55例,非肥胖组59例),采用聚合酶链反应-限制性片断多态性及凝胶电泳方法分析UⅡ基因第4外显子89位点由Ser置换成Asn的变异,即S89N。结果:野生型Ser/Ser基因型频率在ND组为64.9%,T2DM组为52.4%;突变型Asn/Asn基因型频率在ND组为5.3%,T2DM组为16.2%,差异均有统计学意义(P0.05);UⅡ基因型S89N与T2DM有一定关系(P0.01),而与肥胖无明显关系(P0.05)。T2DM组各基因型间空腹葡萄糖、餐后2h血糖、空腹胰岛素、餐后2h胰岛素、胰岛素抵抗指数、胰岛素敏感数、三酰甘油差异均有统计学意义(P0.05~P0.01);空腹C肽和餐后2hC肽在Asn/Asn基因型与其他2种基因型差异均有统计学意义(P0.01);其余指标各基因型间差异均无统计学意义(P0.05)。结论:蚌埠市汉族人群UⅡ基因S89N多态性与T2DM有明显相关性,与肥胖无相关性。

       

      Abstract: Objective:To investigate the association of urotensin Ⅱ(UⅡ)gene S89N polymorphism with type 2 diabetes mellitus(T2DM)and obesity.Methods:One hundred and five patients with T2DM(53 obesity,52 normal)and 114 non-diabetic subjects(55 obesity,59 normal)in Bengbu,Anhui province were randomly selected in the study.The polymerase chain reaction restriction fragment length polymorphism and the gel electrophoretic mobility shift assay were used to examine the S89N polymorphism.Results:The frequency of the Ser/Ser genotype was 64.9% in non-diabetic subjects and 52.4% in T2DM patients.The Asn/Asn frequencies were 5.3% and 16.2% in the two groups respectively.The difference was significant(P0.05).The genotype S89N of UⅡwas related with T2DM(P0.01),but not with obesity(P0.05).The difference of metabolic indexes of FPG,PBG,FINS INS2h,HOMA-Ir,ISI and TG among three genotypes of UⅡ in T2DM group was significant(P0.05-P0.01).The difference of metabolic indexes of CP and CP2h between Asn/Asn and the other two genotypes of UⅡ in T2DM group was significant(P0.01).Conclusions:The UⅡ gene S89N polymorphism is significantly associated with T2DM,but not with obesity among the Han population in Bengbu city.
