姜黄素对氧化型低密度脂蛋白诱导的血管内皮细胞炎症损伤的保护作用

摘要: 目的:研究姜黄素是否可抑制血管内皮的损伤及炎症反应从而起到预防动脉粥样硬化(AS)的作用。方法:培养人脐静脉内皮细胞(HUVEC),加入外源性氧化型低密度脂蛋白(OX-LDL组)组构造与AS发病早期相似的体外模型,用姜黄素干预后(姜黄素组)观察HUVEC形态的改变及炎症因子细胞间黏附分子-1(ICAM-1)、白细胞介素-6(IL-6)、IL-8表达的变化。结果:OX-LDL诱导的AS模型内皮细胞增殖减少,ICAM-1、IL-6、IL-8的表达增加,均高于阴性对照组(P0.01);姜黄素组可减少细胞凋亡,降低ICAM-1、IL-6、IL-8的表达,与OX-LDL组差异均有统计学意义(P0.01)。结论:姜黄素能降低内皮细胞炎症损伤时相关炎症因子的表达,保护血管内皮细胞,预防AS。Abstract: Objective: To study whether curcumin has the protective effect on atherosclerosis via modulation inflammation and endothelial function. Methods: Human umbilical veins endodermis cells(HUVEC) were cultured in vitro. The effects of curcumin on the changes of vascular endothelial cell morphology and the expressions of inflammation factors including inter-cellular adhesion molecule-1(ICAM-1) , interleukin-6 (IL-6) and interleukin-8(IL-8) were studied on the early phase model of atherosclerosis induced by oxidative low-density lipoprotein(OX-LDL) . Results: Compared with control group,OX-LDL inhibited the proliferation of HUVEC and induced the expressions of ICAM-1, IL-6 and IL-8 in HUVEC,which were higher than in control group(P < 0. 01) . Compared with ox-LDL group,curcumin reduced the apoptosis of HUVEC, the expressions of ICAM-1, IL-6 and IL-8 were decreased,which were statistical different(P < 0. 01) . Conclusions: Curcumin can improve atherosclerosis and protect vascular endothelial cells by inhibition of the expressions of inflammatory factors.
