吲哚胺-2,3-双加氧酶在非小细胞肺癌的表达及其意义

    Expression of indoleamine 2,3-dioxygenase in non-small cell lung cancer and its significance

    • 摘要: 目的:探讨非小细胞肺癌(non-small cell lung cancer,NSCLC)组织(肺癌组织及配对淋巴结转移癌组织)中吲哚胺-2,3-双加氧酶(indoleamine-2,3-dioxygenase,INDO)的表达水平与NSCLC临床病理学参数之间的关系。方法:用免疫组织化学检测80例肺癌组织(包括原发性肺癌组织和配对淋巴结转移癌组织)、癌旁正常肺组织石蜡切片中INDO的表达水平,Western blot法检测其中40例新鲜肺癌组织及其癌旁正常肺组织中INDO的表达情况,根据手术、病理和影像学结果判断有无转移并确定分期,分析INDO蛋白表达水平与临床病理学参数之间的关系。结果:INDO蛋白阳性部位位于肺癌细胞质,其在原发性肺癌组织中阳性率为80.00%,明显高于在癌旁正常肺组织中的阳性率20.00%(P<0.001),在有淋巴结转移组中的阳性率为95.56%,显著高于在无淋巴结转移组中的阳性率60.00%(P<0.01),且与淋巴结转移有一定关系(P<0.05)。淋巴结转移癌中INDO阳性表达率为97.78%,显著高于原发灶中INDO的表达率80.00%(P<0.01)。结论:INDO在肺癌原发灶表达显著高于正常肺组织,且与NSCLC的淋巴结转移显著相关,而淋巴结转移癌中的表达又显著高于配对肺癌原发灶中的表达,提示INDO异常高表达可能与NSCLC转移有关。

       

      Abstract: Objective: To assess the relevance of the expression of indoleamine-2,3-Dioxygenase(INDO) in non small cell lung cancer(NSCLC) and the clinical pathological parameters of NSCLC.Methods: Immunohistochemistry method was used to detect the INDO expression in 80 NSCLCs,including primary pulmonary carcinoma and lymph node metastatic carcinoma,and the tissue samples of 40 cases were screened by Western blot analysis to detect the expression of INDO in the cancer and normal tissues.The metastasis stage was defined on the basis of the results of operation,pathological test and imaging diagnosis.The correlation between the expression of INDO and the clinical pathological parameters was analyzed.Results: Immunohistochemistry showed that the INDO was present at the cytoplasm area,and the positive rate was 80.00% in the primary cancer tissue,demonstrating a great increase in comparison with the control(20.00%)(P<0.001),and the positive rate was 95.56% in the metastasis carcinoma group and 60.00% in the none metastasis group(P<0.01).The INDO expression in lymph node metastatic carcinoma was 97.78%,which was significantly higher than that in the primary ones(80.00%)(P<0.01).Conclusions: INDO is over-expressed in the NSCLC group,and is obviously related with the lymph node metastasis of NSCLC,which indicates that over expression of INDO may have a positive correlation with the metastasis of NSCLC.
