    对亚胺培南耐药的肺炎克雷伯菌KPC基因型分析

    Analysis of KPC genotype in imipenem resistant-Klebsiella pneumoniae

    • 摘要: 目的:分析对亚胺培南耐药的肺炎克雷伯菌KPC基因型。方法:使用改良的Hodge实验(MHT)初筛产KPC的肺炎克雷伯菌,PCR和测序鉴定耐药肺炎克雷伯菌是否带有KPC基因,并通过GenBank的比对确定KPC基因型。用脉冲场凝胶电泳(PFGE)技术对KPC阳性菌株进行同源性分析。结果:13株对亚胺培南耐药的肺炎克雷伯菌中有10株MHT结果为阳性,阳性率为76.92%,其中有2株细菌为KPC基因阳性,阳性率为15.38%,通过GenBank比对,此KPC基因为2型。PFGE结果显示这2株细菌来源于同一克隆。结论:分离出了携带有KPC-2型碳青霉烯酶基因的肺炎克雷伯菌。

       

      Abstract: Objective: To analyze the KPC genotype in imipenem resistant-Klebsiella pneumoniae. Methods: The modified Hodge test (MHT) was used to screen the KPC enzyme-producing Klebsiella pneumoniae; the KPC gene was confirmed by PCR and DNA sequencing, and the KPC genotype was further confirmed by comparison with GenBank from internet. Homology of KPC was analyzed by using pulsed-field gel electrophoresis(PFGE) . Results: Ten of the 13 strains(76. 92%) of imipenem resistant-Klebsiella pneumoniae were proved positive by MHT,2 (15. 38%) of which presented positive KPC gene; and the KPC gene was confirmed as type-2 by comparision with GenBank. The Results of PFGE demonstrated that the 2 strains came from the same clone. Conclusions: KPC-2 type carbapenemases gene in Klebsiella pneumoniae has been isolated.
