肿瘤坏死因子-对体外培养成骨细胞一氧化氮水平的影响

    Effect of tumor necrosis factor- on the nitric oxide level in cultured osteoblasts in vitro

    • 摘要: 目的:检测成骨细胞在不同质量浓度肿瘤坏死因子-(TNF-)作用下一氧化氮(NO)的水平。方法:培养及鉴定成骨细胞,检测不同质量浓度TNF-作用下成骨细胞培养上清液中NO的含量。结果:随着TNF-质量浓度的提高,NO的水平逐渐升高;当TNF-为10g/L时NO水平最高,此后随着TNF-作用时间和质量浓度的增加NO水平减少(P0.05~P0.01)。结论:单一的TNF-可以诱导成骨细胞产生NO,但NO并不随着TNF-质量浓度和时间的增加而持续增多。

       

      Abstract: Objective: To explore the level of nitric oxide(NO) in osteoblasts treated with different concentrations of tumor necrosis factor-(TNF-) . Methods: The osteoblasts were cultured and identified. The NO level in cultured supernafants in osteoblasts treated with different concentrations of TNF- was measured. results: The level of the NO was increased with the increase of concentration of TNF-,which was the maximum at 10 g /L of TNF-. The level of the NO began to decrease with the increase of processing time and concentration of TNF-(P < 0. 05 to P < 0. 01) . Conclusions: TNF- can induce the expression of NO,but TNF- can not countine to increase the expression of NO with the increase of its processing time and concentration in cultured osteoblasts.
