    The relevant research between expression of ribonuclease inhibitor and the malignant proliferation in human breast cancer cells

    • 摘要: 目的:探讨核糖核酸酶抑制因子(ribonuclesae inhibitor,RI)的表达与人乳腺癌细胞MCF-7、MDA-MB-231、SKBR-3恶性增殖之间的关系。方法:四甲基偶氮唑蓝(MTT)法检测细胞增殖情况,流式细胞术检测细胞周期分布,RT-PCR检测RI基因表达水平,细胞免疫化学技术检测RI蛋白表达情况。结果:人乳腺癌细胞MCF-7细胞增殖速度、S期所占比例均小于人乳腺癌细胞MDA-MB-231、SKBR-3(P<0.05~P<0.01),且MCF-7细胞中RI基因表达水平、蛋白表达水平明显高于MDA-MB-231、SKBR-3细胞(P<0.01)。结论:RI与人乳腺癌细胞恶性增殖生长有一定的相关性。

       

      Abstract: Objective: To study the relation between the change of ribonuclease inhibitor(RI) and malignant proferations in different malignant degree human breast cancer cell lines MCF-7、MDA-MB-231、SKBR-3.Methods: Cell profigeration was determined by MTT assay;The cell cycle was measured by flow cytometry(FCM);the expression of RI mRNA was examined by semi-quantitative RT-PCR;immunohistochemistry was used to detected the protein expression of RI.Results: Human breast cancer cell lines MCF-7 cell malignant proliferation.S period was less than the propotion of human breast cancer cell lines MDA-MB-231,SKBR-3(P<0.05-P<0.01),and the expressions of RI mRNA and protein in the human breast cancer cell lines MCF-7 were higer than that of human breast cancer cell lines MDA-MB-231、SKBR-3(P<0.01).Conclusions: RI and human breast cancer cells malignant profigeration growth has certain relevance.
