    The experimental study of kidney oxidation damage induced by trichloroethylene in rats

    • 摘要: 目的: 探讨三氯乙烯(trichloroethylene,TCE)对大鼠肾脏组织的脂质过氧化损伤效应及一氧化氮(NO)表达的影响。方法: 32只大鼠随机分为对照组和500、1000、2000 mg/kg TCE三个染毒组,每组8只,每天1次,连续7d皮下注射染毒,染毒结束后计算肾脏脏器系数,制备肾组织匀浆,测定超氧化物歧化酶(SOD)、丙二醛(MDA)、NO的含量。结果: 染毒第4天大鼠体重增加量,500 mg/kg组均高于1000 mg/kg组和2000 mg/kg组(P<0.05和P<0.01);各组肾脏及脏器系数差异均无统计学意义(P>0.05)。与对照组比较,肾组织匀浆中SOD活力500、1 000、2 000 mg/kg TCE染毒组均低于对照组(P<0.01和P<0.05);TCE 3个染毒组中MDA含量均高于对照组(P<0.05~P<0.01);500、1 000和2 000 mg/kg TCE染毒组NO水平均高于对照组(P<0.05~P<0.01)。结论: 三氯乙烯对肾脏有一定的损伤作用,使肾脂质过氧化作用增强。

       

      Abstract: Objective: To explore the effects of trichloroethylene(TCE)on kidney peroxidation damage and the expression of nitric oxide(NO)of rats.Methods: Thirty-two SD rats were randomly divided into the control group and low,medium and high dose TCE injection group(8 rats per group),which were subcutaneously injected with olive oil and 500,1000 and 2000 mgkgP>-1P>d P>-1P>TCE respectively once a day for 7 days.The kidney index and activities of superoxide dismutase(SOD)and contents of malondialdehyde (MDA)and NO of kidney tissue were measured.Results: The weight of rats with TCE injection increased on the fourth day, the increasing weight of low dose group were higher than that of medium and high dose group(P<0.05 and P<0.01)and the kidney index was not statistical significant(P>0.05).The activities of SOD of three different dose groups were lower than that of the control group(P<0.01 and P<0.05),but for the concentration of MDA and levels of NO of three different dose groups were higher than that of the control group(P<0.05-P<0.01).Conclusions: TCE has certain injury effects on kidney tissue,which can strengthen the effects of renal lipid peroxidation.
