    The expression and relevance of PTEN and p53 in hepatocellular carcinoma

    • 摘要: 目的:探讨肝细胞癌(hepatocellular carcinoma,HCC)第10号染色体缺失的磷酸酶张力蛋白同源物基因(gene of phosphate and tension homology deleted on chromsome ten,PTEN)和p53蛋白的表达及其临床意义。方法:采用免疫组织化学EliVision方法测定70例HCC组织和40例相应的癌旁组织中PTEN和p53蛋白的表达,分析两个指标与临床病理特征之间的关系以及它们之间的相关性。结果:PTEN在HCC组织中的阳性率为57.1%,显著低于癌旁组织的95.0%(P0.01)。在肿瘤分期晚和有淋巴结转移的患者中,PTEN表达均降低(P0.01)。p53蛋白在HCC组织中表达率为60.0%,显著高于癌旁组织的15.0%(P0.01)。在肿瘤大小、病理分级、TNM分期和淋巴结转移等临床病理指标上,p53蛋白表达差异均无统计学意义(P0.05)。PTEN与p53蛋白表达在HCC组织中呈负相关关系(P0.05)。结论:PTEN低表达与HCC进展有关,有可能作为评价肝癌预后的指标。

       

      Abstract: Objective: To investigate the expression and clinical significance of gene of phosphate and tension homology deleted on chromsome ten( PTEN) and p53 in hepatocellular carcinoma( HCC) sample. Methods: The protein levels of PTEN and p53 were examined by EliVision immunohistochemistry in 70 samples of HCC tissue and 40 samples of adjacent hepatic tissue,while the correlation between the expression and clinicopathological features was analyzed. Results: The positive expression rate of PTEN in HCC tissue was 57. 1%,significantly lower than that in adjacent hepatic tissue,which the positive expression rate was 95. 0%( P 0. 01). Patients with advanced stage of tumor or lymph node metastasis had a lower tendency to express PTEN in HCC tissue( P 0. 01). The positive expression rate of p53 was 60. 0% in HCC tissue,significantly higher than that in adjacent hepatic tissue,which the positive expression rate was 15. 0%( P 0. 01). There were no statistical significance for the p53 expression in HCC tissue in clinicopathological parameters such as tumor size,pathologic grade,TNM stage and lymphnode metastasis( P 0. 05). The expression of PTEN was negatively related with that of p53 in HCC tissue( P 0. 05). Conclusions: The expression of PTEN may be associated with the progression of HCC. It may serve as an index for prognosis of HCC.
