Abstract:
Objective: To observe the influence of glucose on the expression of Apolipoprotein E(Apo E) in human monocytic leukemia cell line(THP-1) macrophages.
Methods: THP-1 cells were induced to macrophages with phorbol,and then treated with RPMI 1640 culture medium containing diferent concentrations of glucose for 72 hours.Apo E mRNA and protein level were analyzed by RT-PCR and Western Blot respectively,meanwhile the viability of macrophages was assayed with MTT method.
Results: Glucose significantly decreased Apo E expression of THP-1 macrophages in a dose-dependent manner(20-30 mmol/L).
Conclusions: Glucose decreases Apo E expression of THP-1 macrophages,which might be one of the mechanisms of proatherosclerosis of glucose.
