Abstract:
Objective: To investigate the effect of genistein on the apoptosis of cerebellar neuronal cells induced by acrylamide.
Methods: The model of cerebellar neuronal apoptosis in rats was established by intragastric administration of acrylamide,and their independence behaviour was observed,the index of oxidative stress:GSH-PX,SOD and MDA was determined,the changes of cerebellar neuronal cells was observed electron microscopy.In the meantime,the protective effect of Einstein was observed.
Results: The frequency of voluntary movements in rats of the group treated with acrylamide was significantly lower than that of control group(
P<0.01).The contents of GAH-PX,SOD of cerebellar tissue of the group treated with acrylamide were significantly lower than that of control group(
P<0.01) and the contents of MDA was much higher than that of control group(
P<0.01).The edema of cerebellar cells,the vacuolar degeneration of plastochondria and the apoptotic bodies were observed under electron microscope.Treatment with 25 mg·kg
-1·d
-1 and 50 mg·kg
-1·d
-1 of genistein increased the contents of GAH-PX and SOD,and reduced the content of MDA and the injury of cerebellar cells.
Conclusions: The results indicate that genistein can reduce cerebellar neurocyte apoptosis induced by acrylamide.
