Abstract:
Objective:To investigate the effect of angiotensin Ⅱ(AngⅡ) on fibronectin(FN) biosynthesis on human renal tubular epithelial cells.
Methods:Cultured human renal tubular epithelial cells were treated with AngⅡ at the concentrations of 10
-6 mol/L for 6,12,24,48,72 and 96 hours;and the cells were treated with AngⅡ at the concentrations of 10
-8,10
-7 and 10
-6 mol/L for 96 hours. The FN expression in renal tubular cells was determined by both Western blot and immunofluorescence.
Results:Fibronectin synthesis was increased by AngⅡ at a concentration of 10
-6 mol/L in the renal tubular epithelial cells. The expression of FN started to increase after 12 hours, and reached the peak after 72 hours. The synthesis of FN was the most obvious at a concentration of 10
-6 mol/L,and the following was at 10
-7 and 10
-8 mol/L.
Conclusions:It suggests that AngⅡ stimulates FN secretion of human renal tubular epithelial cells in a dose and time-dependent manner.
