γ-干扰素在体外对血管内皮细胞凋亡的诱导作用

    Interferon-γinduces the apoptosis of vascular endothelial cell in vitro

    • 摘要: 目的: 观察γ-干扰素(IFN-γ)对血管内皮细胞(EC)的作用及其对肿瘤坏死因子-α(TNFα)表达的影响。方法: 体外培养EC,MTT法检测IFN-γ(0、20、40、80μg/L)对EC生长的直接影响,半定量逆转录-多聚酶链反应检测不同浓度的IFN-γ对EC凋亡相关基因TNFαmRNA的表达的影响。结果: IFN-γ可抑制体外培养的EC生长(P<0.01),且具有剂量依赖;IFN-γ能明显上调EC的TNFα表达。结论: IFN-γ可能通过上调EC中TNFα的表达而诱导EC凋亡。

       

      Abstract: Objective: To explore the role of interferon-γ(IFN-γ) in inducing the apoptosis of vascular endothelial cells(EC) in vitro and its influence on the expression of TNFα.Methods: The EC cells were cultured in vitro with the condition media containing varied concentration of IFN-γ(0,20,40,80 μg/L) for 24 hours,and then the total RNA was extracted to analyze the mRNA level of TNFα in EC by RT-PCR.The influence of IFN-γ in inducing the apoptosis on EC was analyzed by MTT method.Results: IFN-γ could induce the apoptosis of EC in a dose-dependent manner and it could upregulate the expression of TNFα mRNA level in EC.Conclusions: IFN-γ can induce the apoptosis of EC in vitro by up-regulating the expression of TNFα.
