三氧化二砷抑制恶性黑色素瘤B16细胞核酸合成的研究

    Effects of arsenic trioxide on nucleic acid biosynthesis and apoptosis of melanoma B16 cells

    • 摘要: 目的: 研究不同浓度的三氧化二砷(As2O3)对恶性黑色素瘤B16细胞核酸合成抑制及诱导B16细胞凋亡的作用,探讨砷及其化合物治疗恶性黑色素瘤的作用机制,为其治疗恶性黑色素瘤提供新的理论和实验依据。方法: 采用3H-TdR和3H-UdR掺入肿瘤细胞DNA和RNA的方法,观察不同浓度的As2O3对恶性黑色素瘤B16细胞核酸生物合成的抑制作用;用流式细胞术(FCM)检测As2O3诱导B16细胞的凋亡及细胞毒作用。结果: As2O3对恶性黑色素瘤B16细胞核酸合成抑制和诱导凋亡及细胞毒作用有显著的浓度和时间依赖关系(P<0.01)。结论: As2O3对恶性黑色素瘤B16细胞核酸合成的抑制作用和诱导凋亡及细胞毒作用随药物浓度的增加和作用时间的延长而增强。

       

      Abstract: Objective: To study the effect of arsenic trioxide(As2O3) on nucleic acid biosynthesis and apoptosis of melanoma B16 cells.Methods: The nucleic acid biosynthesis in melanoma B16 cells were detected by incorporation of 3H-TdR and 3H-UdR into DNA and RNA,respectively.Apoptosis and necrosis in B16 cells were analyzed by flow cytomertry.Results: As treated with different doses of As2O3 in B16 cells the rates of inhibition and apoptosis and necrosis have both dose-dependent and time-dependent manner(P<0.01).Conclusions: As2O3 can inhibit nucleic acid biosynthesis and induce apoptosis and necrosis in melanoma B16 cells.The assay showed a dose-response and time-response relationship for inhibition of nucleic acid biosynthesis and apoptosis in B16 cells by As2O3.The inhibition was enhanced with the increase of As2O3 concentration and prolong of co-culture time.
