    Decreasing toxicity and synergistic effects of total glucosides of paeony on tumor-bearing mice treated by cyclophosphamide

    • 摘要: 目的: 探讨白芍总苷(total glucosides of paeony,TGP)对环磷酰胺(CTX)治疗荷瘤小鼠的增效减毒作用,为寻找有效低毒的抗肿瘤药物及提高肿瘤化疗中的生存率提供有价值的实验资料。方法: 建立鼠S180腹水瘤及实体瘤模型,分别给予生理盐水(NS组)、不同剂量的TGP (TGP组)、CTX (CTX组)及CTX+TGP (TGP+MTX组)处理,连续给药10 d,观察腹水瘤小鼠的生存时间,计算生命延长率;检测实体瘤组瘤重、脾重及胸腺重,计算抑瘤率、脾指数及胸腺指数,并对T-淋巴细胞转化率及血清肿瘤坏死因子(TNF-α)水平进行检测。结果: 与NS组比较,100 mg/kg和200 mg/kg TGP均可延长荷瘤小鼠的存活时间,差异有统计学意义(P < 0.01),且对S180移植肿瘤均有抑制作用,其抑瘤率分别为18%和30%。TGP组小鼠体重及脏器指数均高于NS组(P < 0.01)。TGP可增加T-淋巴细胞的转化率,升高血清TNF-α水平(P < 0.01)。结论: TGP对小鼠S180腹水瘤及实体瘤的生长具有一定的抑制作用,并可增加荷瘤小鼠的免疫功能,增强CTX的抗肿瘤作用,降低其对机体的毒性作用。

       

      Abstract: Objective: To study the toxicity-decreasing and synergistic effect of total glucosides of paeony(TGP) on tumor-bearing mice treated by cyclophosphamide(CTX) for providing some valuable experimental data for searching high efficiency and low toxicity antitumor drugs and improving survival rate. Methods: S180 tumor cells were inoculated into healthy mouse to construct the S180 bearing-mice model,and treated with normal saline,TGP,CTX and TGP plus CTX respectively for 10 days,Observed live time to evaluate life extension rate for ascitic tumor,and measured tumor,spleen,and thymus weight to evaluate anti-tumor rate,index of spleen and thymus. Further more, lymphocyte proliferation rate and the plasma levels of tumor necrosis factor-α (TNF-α) were detected. Results: The high dose TGP significantly increased the survival time of mice bearing sarcosarcoma sarcoma S180 when compared with mice treated with normal saline,and decreased tumor weight and increased indexes of immune organs. TGP can strengthen anti-tumor activity of CTX,antagonize the inhibition of CTX on the body and immunologic system,and improve lymphocyte transformation and TNF-α level in plasma. Conclusions: TGP has inhibitory effects on the growth of tumor in S180 sarcoma-bearing mice and TGP can increase the activity of body immunity,improve the anti-tumor effects of CTX,and attenuate the toxicity of CTX as well.
