    链脲佐菌素诱导大鼠糖尿病肾病模型的建立及肾脏病理结构改变

    Pathological structural changes of the kidney in streptozotocin-induced rat model of diabetic nephropathy

    • 摘要: 目的:探讨糖尿病大鼠肾脏结构的变化,建立糖尿病肾病大鼠模型。方法:雄性清洁SD大鼠48只,随机分为实验组(DM组,n=24)和对照组(NC组,n=24)。DM组大鼠一次足量腹腔注射链脲佐菌素(STZ)制备糖尿病动物模型,NC组给予等量的枸橼酸-枸橼酸钠缓冲液。分别于注射后1、4、8和12周,检测体重、血糖、24 h尿量、24 h尿蛋白、肾重指数(肾脏重量/体重)和肾脏病理结构的变化。结果:与NC组相比,DM组大鼠血糖明显升高,肾重指数增加(P<0.01);随着病程的延长,24 h尿量、24 h尿蛋白量均持续升高(P<0.01)。病理切片显示,DM组大鼠肾脏重量增加、肾小体肿胀、肾小囊腔缩小、血管球内皮细胞排列紊乱和肾小体基膜增厚等。结论:STZ可诱导糖尿病大鼠肾脏结构和功能受损,成功制备糖尿病肾病大鼠模型。

       

      Abstract: Objective:To explore pathological structural changes in the kidney of diabetic rats and establish the rat model of diabetic nephropathy.Methods:Forty eight male Sprague-Dawley rats were randomly divided into control group and experimental group (24 for each).Twenty-four rats in the experiment group were intraperitoneally injected with streptozotocin to set up animal modal of diabetes.At 1,4,8 and 12 weeks,the were recovded weight,blood glucose,24-hour urine volume,24-hour urinary protein,kidney hypertrophy index and pathological changes of kidney.Results:The diabetic group was detected.The higher level (P<0.01) of blood glucose,more 24 hour urine and 24 hour urinary protein than the control group (P<0.01).The changes of kidney structure was obvious in light microscope.Conclusions:Kidney damage of diabetic rats had been induced by streptozotocin injection,and the rat model of diabetic was successively established.
