毛细管电泳-激光诱导荧光检测大鼠脑组织多巴胺含量

    Determination of dopamine in the brain tissue of rats by using capillary electrophoresis with laser-induced fluorescence method

    • 摘要: 目的: 采用毛细管电泳-激光诱导荧光法建立检测大鼠脑组织多巴胺含量的方法。方法: 低温匀浆大鼠脑组织离心后,取上清用pH11.010mmol/L硼砂缓冲液,在室温下衍生反应10h连接上FITC荧光基团;毛细管电泳分离条件为pH10.3100mmol/L硼酸-Tris缓冲液。结果: 在选定的实验条件下,方法检测限(S/N=3)达2.1nmol/L水平,批内相对标准差(RSD)为3.4%,回收率达96.05%,在10-8~10-10mol/L范围内呈良好线性关系。结论: 应用毛细管电泳-激光诱导荧光法可检测鼠脑组织多巴胺含量。

       

      Abstract: Objective: To establish a method of determining the dopamine in the brain tissue of rats by using capillary electrophoresis with laser-induced fluorescence.Methods: The homogenate of the rats' brain tissue was centrifugated in low temperature and the supernatant was reacted in pH 11.0 10 mmol/L borax buffer solution.The reaction of derivatization time was 10 hours under alkaline (10 mmol/L Na2B4O7 pH 11.0);mobile condition of capillary electrophoretic separation was a mixture of 100 mmol/L Boric acid-Tris.Results: Detection limits of the method were determined to be lower than 2.1 nmol/L and the RSD values were 3.4%;the recoveries were above 95.6%.The linearity was from 10-10 to 10-8 mol/L.Conclusions: The method of detecting dopamine in the brain of rats is convenient by using capillary electrophoresis with laser-induced fluorescence.
