Abstract:
Objective: To establish a method of determining the dopamine in the brain tissue of rats by using capillary electrophoresis with laser-induced fluorescence.
Methods: The homogenate of the rats' brain tissue was centrifugated in low temperature and the supernatant was reacted in pH 11.0 10 mmol/L borax buffer solution.The reaction of derivatization time was 10 hours under alkaline (10 mmol/L Na

7 pH 11.0);mobile condition of capillary electrophoretic separation was a mixture of 100 mmol/L Boric acid-Tris.
Results: Detection limits of the method were determined to be lower than 2.1 nmol/L and the RSD values were 3.4%;the recoveries were above 95.6%.The linearity was from 10
-10 to 10
-8 mol/L.
Conclusions: The method of detecting dopamine in the brain of rats is convenient by using capillary electrophoresis with laser-induced fluorescence.
