全反式维A酸诱导肺癌细胞株凋亡的研究

    Apoptosis of lung cancer cell line induced by all-trans-retinoic acid

    • 摘要: 目的: 探讨全反式维A酸(ATRA)对小细胞肺癌细胞株SH-77的治疗作用及机制,寻找肺癌治疗的新途径。方法: 采用不同浓度的ATRA处理肺癌细胞株SH-77,通过苔盼蓝拒染实验检测细胞活力、MTT法检测细胞生长抑制率和流式细胞术检测细胞周期分布、细胞凋亡和Bcl-2蛋白的表达。结果: 不同浓度的ATRA对SH-77的生长均有抑制作用,G1/G0期细胞比例升高,S期细胞比例降低,增殖指数(PI)降低,凋亡指数(AI)和AI/PI比值升高,诱导细胞凋亡;Bcl-2蛋白比例下降(P<0.01)。结论: ATRA对肺癌细胞株SH-77的生长有抑制作用,同时可诱导其凋亡,可能与下调其Bcl-2蛋白表达有关。

       

      Abstract: Objective: To investigate the effect and mechanism of all-trans-retinoic acid(ATRA) in inducing apoptosis of human small cell lung cancer(SCLC) cell line SH-77.Methods: SCLC cell line SH-77 was treated with different concentrations of ATRA.The multiparameter such as cell growth inhibition rate,proliferation index(PI),apoptosis index(AI) and Bcl-2 protein expression was detected by Typan blue stainning,MTT and flow cytometry,respectively.Results: Different concentrations of ATRA could inhibit cell line SH-77 cell growth.G1/G0 stage and AI were increased,while cell count at S stage and PI were reduced.Bcl-2 protein expression was decreased (P<0.01).Conclusions: ATRA can inhibit SCLC cell lines SH-77 growth,induce cell apoptosis and reduce Bcl-2 protein expression.
