营养性幼年肥胖雌性大鼠膈肌收缩功能与超微结构的观察

    The changes of diaphragm contractility and ultrastructure in diet-induced obesity impuberism female rat

    • 摘要: 目的: 观察营养性肥胖幼年雌性大鼠的膈肌收缩功能及膈肌超微结构的变化,探讨可能的发生机制。方法: 将30只雌性刚离乳SD大鼠按体重随机分成模型组和对照组各15只,对照组给予普通饲料喂养,模型组给予改良的高脂饲料喂养;喂养8周后测定2组大鼠的体重,并应用体外灌流大鼠膈肌条的方法,测定其膈肌单收缩张力(Pt)、最大强直张力(Po)、张力-频率关系及疲劳指数(FI)的变化,同时电镜观察膈肌组织的形态和超微结构的变化。结果: 模型组大鼠膈肌Pt、Po和FI值均低于对照组(P < 0.05~P < 0.01);在给予10、20、40、60、100 Hz的刺激下,模型组膈肌条张力均明显低于对照组(P < 0.01);电镜下模型组大鼠膈肌可见大量脂滴分布;肌纤维明显肿胀,少数肌丝紊乱;线粒体数量减少,部分空泡变,嵴模糊,见髓样小体样改变。结论: 高脂饮食诱导的幼年大鼠营养性肥胖,膈肌收缩功能减弱,易疲劳,可能与其膈肌组织脂质沉积增加,线粒体破坏有关。

       

      Abstract: Objective: To measure the changes of diaphragm contracting function and ultrastructure in diet-induced obesity impuberism female Sprague-Dawley rats,and explore the possible mechanism. Methods: Thirty female Sprague-Dawley weaning rats were randomly divided into model group and control group according to their weight. The rats in control group were fed with standard chow and in model group were fed with high fat diet for 8 weeks. Body weight was compared between the both groups at the end of experiment. In perfusion diaphragm strips model in vitro,the parameters which reflex the diaphragm contractility including peak twitch tension (Pt),maximum tetanic tension (Po),force-frequency relationship and fatigue index (FI) were assessed by Medlab-U/4C biological signal collecting system. The changes of diaphragm ultrastructure were studied by electron microscopy in rats. Results: Compared with the rats in control group,Pt,Po,FI were lower in model group (P < 0.05-P < 0.01). Compared with the rats in control group,tetanic force under the stimulus frequency of 10,20,40,60,100 Hz were decreased in model group (P < 0.01). Electron microscope morphometry of diaphragm in model group revealed a large number of lipid droplets was distributed in diaphragm,muscle fiber edema,and small number of filaments disorder were observed;the quantity of mitochondria was decreased,mitochondria was oedema and expanded,its cristae broken and vague,a great quantity of mitochondria were vacuolization or vesiculation,and myelin figures was observed. Conclusions: In dietinduced obesity Sprague-Dawley rats,the contractile function of diaphragm is decreased and likely to fatigue. The mechanism may be related to fat deposits in diaphragm and breakage of mitochondria.
