    孕鼠接触葡萄球菌肠毒素B对子代新生鼠TCR Vβ8+T细胞的影响

    Effect of maternal SEB administration on the TCR Vβ8+T cells of the offspring newborn rats

    • 摘要: 目的:观察孕鼠接触葡萄球菌肠毒素B(SEB)对其子代新生鼠胸腺及外周血 TCR Vβ8+ T细胞的影响。方法:在妊娠16 d时给予SD大鼠尾静脉注射15 μg SEB(SEB组),同时设立注射磷酸盐缓冲溶液对照组。孕鼠自然分娩后获取出生后0~5 d的子代新生鼠胸腺及外周血,流式细胞仪检测其TCR Vβ8+ T细胞比例。结果:与对照组比较,SEB组出生后0~3 d的新生鼠胸腺中CD4+Vβ8+ T细胞比例均减少(P<0.05~P<0.01),出生后4~5 d 2组差异均无统计学意义(P>0.05);出生后0~5 d,SEB组胸腺中CD8+Vβ8+ T细胞比例均低于对照组(P<0.05~P<0.01);出生后0~5 d,SEB组CD4+Vβ8+及CD8+Vβ8+ T细胞的绝对数均较对照组明显减少(P<0.01)。新生鼠出生后0~5 d,SEB组外周血CD4+Vβ8+ T细胞比例均较对照组明显减少(P<0.01);CD8+Vβ8+ T细胞比例亦均较对照组减少(P<0.05~P<0.01);出生后0~5 d,SEB组外周血CD4+Vβ8+及CD8+Vβ8+ T细胞绝对数均较对照组明显减少(P<0.01)。结论:妊娠期大鼠接触SEB可减少子代新生鼠胸腺及外周血TCR Vβ8+ T细胞,并保留至成年期。

       

      Abstract: Objective:To investigate the effects of maternal SEB administration on the TCR Vβ8+T cells in the thymus and peripheral blood of offspring newborn rats.Methods:Pregnant rats with gestational 16 days were injected with 15 μg SEB in 0.2 mL PBS by iv(SEB group),and the control group were treated with the same volume of PBS.The thymus and peripheral blood of newborn offspring rats after 0 to 5 days of delivery were harvested,and the levels of TCR Vβ8+ T cells of whose were analyzed using the flow cytometry.Results:Compared with the control group,the percentage of CD4+ TCR Vβ8+T cells in thymus of offspring newborn rats after 0 to 3 days of delivery significantly decreased in SEB group(P<0.05 to P<0.01),and the difference of which in thymus of offspring newborn rats after 4 to 5 days of delivery between two groups was not statistically significnat(P>0.05).In the thymus of offspring newborn rats after 0 to 5 days of delivery,the percentage of CD8+ TCR Vβ8+ T cells in SEB group was significantly lower than that in control group(P<0.05 to P<0.01),and the absolute number of CD4+Vβ8+ and CD8+Vβ8+ T cells in SEB group significantly decreased compared with the control group(P<0.01).In the peripheral blood of offspring newborn rats after 0 to 5 days of delivery,the percentages of the CD4+ Vβ8+ and CD8+ Vβ8+ T cells,and the absolute number of CD4+ Vβ8+ and CD8+ Vβ8+ T cells in SEB group significantly decreased compared with the control group(P<0.05 to P<0.01).Conclusions:Maternal SEB administration during pregnancy can decrease the levels of TCR Vβ8+T cells in the thymus and peripheral blood of offspring newborn rats,and which can continue into adulthood.
