    杏仁核点燃大鼠癫痫模型中P-糖蛋白和多药耐药相关蛋白1的动态表达变化

    The dynamic expressions of P-glycoprotein and multidrug resistance protein 1 in amygdala-kindled epileptic model of rat

    • 摘要: 目的:探讨杏仁核点燃大鼠癫痫模型中P-糖蛋白(PGP)和多药耐药相关蛋白1(MRP1)的动态表达变化。方法:选择40只成年雄性大鼠,随机分为模型组和对照组,各20只。对照组予电极植入,不予刺激;模型组建立杏仁核点燃大鼠癫痫模型。制作2组大鼠的脑组织标本,并进行免疫组织化学染色,计算2组大鼠皮层及海马区域24 h、7 d、30 d时的PGP及MRP1阳性细胞数。采取RT-PCR法检测mRNA表达,比较2组大鼠皮层和海马区域24 h、7 d、30 d的PGP mRNA及MRP1 mRNA表达情况。结果:模型制作成功后24 h、7 d和30 d,模型组大鼠皮层及海马区PGP和MRP1阳性细胞数均高于对照组(P<0.05~P<0.01),PGP mRNA及MRP1 mRNA表达亦均高于对照组(P<0.05~P<0.01);模型组组内比较显示,模型制作成功后30 d的大鼠大脑皮层和海马区 PGP、MRP1阳性细胞数和PGP mRNA及MRP1 mRNA表达均高于24 h(P<0.05);而对照组以上指标在各时间点差异均无统计学意义(P>0.05)。结论:在杏仁核点燃模型中,大鼠大脑皮层及海马区的PGP及MRP1表达均增多,且随着时间延长,模型组内表达呈增高趋势。

       

      Abstract: Objective: To investigate the dynamic expressions of P-glycoprotein(PGP) and multidrug resistance protein 1(MRP1) in amygdala-kindled epileptic model of rat.Methods: Forty adult male rats were randomly divided into the model group and control group(20 rats each group).The amygdala-kindled seizure model was established in model group,and the electrode was implanted into the control group without stimulation.The brain tissue samples in two groups were harvested,and detected using immunohistochemistry method.At 24 h,7 d and 30 d of establishing model,the numbers of PGP and MRP1-positive cells in cortex and hippocampus of two groups were calculated,and the mRNA expressions of PGP and MRP1 mRNA in cortex and hippocampus of two groups were detected using RT-PCR.Results: After 24 h,7 d and 30 d of establishing model,the positive cells and mRNA expressions of PGP and MRP1 in cortex and hippocampus in model group were higher than those in control group(P<0.05 to P<0.01).The positive cells and mRNA expressions of PGP and MRP1 in cortex and hippocampus after 30 d of establishing model were higher than those at 24 h of establishing model in model group(P<0.05),but the difference of which at different time-points in control group was not statistically significant(P>0.05).Conclusions: The expession of PGP and MRP1 increases in cortex and hippocampus of amygdala-kindled epileptic rat,with the lengthening of time,it gradually increases in amygdala-kindled epileptic model.
