    Effect of TSA on the expression of type Ⅰ Collagen in the scarring cytology model after glaucoma filtering surgery

    • 摘要: 目的:研究曲古抑菌素A(TSA)对青光眼滤过术(GFS)术后滤过道瘢痕化细胞模型Ⅰ型胶原蛋白表达的影响。方法:构建GFS术后滤过道瘢痕化细胞模型,RT-PCR、Western blotting法分别检测600 nmol/L TSA对组蛋白乙酰基转移酶Ⅰ型胶原蛋白mRNA及蛋白表达的影响。结果:RT-PCR、Western blotting均证实600 nmol/L TSA对Ⅰ型胶原蛋白在基因转录及蛋白表达水平有抑制作用(P<0.01和P<0.05)。结论:TSA能减弱GFS术后滤过道瘢痕化细胞模型中Ⅰ型胶原纤维基因转录及蛋白表达水平。

       

      Abstract: Objective:To investigate the effects of trichostatin A(TSA) on the expression of type Ⅰ Collagen in the scarring cytology model after glaucoma filtering surgery(GFS).Methods:The cytology model of scarring after GFS was established.The effects of 600 nmol/L TSA on the expression levels of type Ⅰ Collagen mRNA and protein were detected using RT-PCR and Western blotting,respectively.Results:The results of RT-PCR and Western blotting showed that the 600 nmol/L TSA could inhibit the expression levels of type Ⅰ Collagen mRNA and protein(P<0.01 and P<0.05).Conclusions:TSA can reduce the expression levels of type Ⅰ Collagen mRNA and protein in the scarring cytology model after GFS.
