    Expression of globular C1q receptor in spontaneous abortion,and its biological significance

    • 摘要: 目的:探索球状C1q受体(gC1qR)在自然流产中的表达及其生物学意义。方法:收集自然流产和人工流产蜕膜组织各30例,以滋养层细胞系HTR-8/SVneo为研究对象,免疫组织化学SP法检测gC1qR蛋白在蜕膜组织中的表达及定位情况;免疫印迹技术及实时荧光定量聚合酶链反应分别检测gC1qR蛋白及mRNA的相对表达水平;流式细胞技术对滋养层细胞凋亡情况进行评估,透射电镜对滋养层细胞凋亡形态进行观察。结果:gC1qR蛋白及mRNA在自然流产蜕膜组织中的表达明显高于人工流产的蜕膜组织(P<0.01);过表达gC1qR基因可明显增加滋养层细胞的凋亡率(P<0.01),且电镜下呈现典型的凋亡形态学改变。结论:gC1qR基因与自然流产的发生紧密相关,有望成为诊断、治疗及预防自然流产的潜在靶点。

       

      Abstract: Objective:To explore the expression of globular C1q receptor(gC1qR) and its biological significance.Methods:Thirty decidual tissue specimen of spontaneous abortion and 30 samples of induced abortion were collected,and extravillous cytotrophoblasts (EVCT)-derived transformed cell lines HTR-8/Svneo was investigated.The protein expression and location of gC1qR in decidual tissue were detected using immunohistochemical SP method,the expression levels of gC1qR protein and mRNA were detected using immunofluorescent staining and qRT-PCR,the apoptosis of trophoblast cells was evaluated using flow cytometry and electronic microscope.Results:The expression levels of gC1qR protein and mRNA in decidual tissue in spontaneous abortion patients were significantly higher than those in induced abortion patients(P<0.01).The overexpression of gC1qR could significantly increase the apoptosis rate of trophoblast cells(P<0.01),and the typical apoptosis morphological change was observed under electronic microscope.Conclusions:The gC1qR gene is closely related to spontaneous abortion,which is maybe to be an potential target point in the diagnosis,treatment and prevention of spontaneous abortion.
