    Sensitization effect of ginsenoside-Rh2 on the apoptosis of human gastric cancer SGC-7901 side population cells induced by low concentration of oxaliplatin

    • 摘要: 目的:探讨人参皂苷Rh2(GS-Rh2)是否可以提高胃癌细胞对奥沙利铂(L-OHP)药物作用的敏感性。方法:使用流式细胞仪检测并分选胃癌SGC-7901的侧群(SP)细胞,使用CCK-8试剂盒检测并验证SP细胞的高增殖能力,使用MTT法检测GS-Rh2和L-OHP对SGC-7901-SP细胞的半数抑制率(IC50),通过流式细胞仪检测GS-Rh2联合低浓度的L-OHP干预后SGC-7901-SP细胞周期及凋亡率的变化,使用Western blotting检测GS-Rh2和L-OHP干预后SGC-7901-SP细胞的凋亡蛋白Bax、Bcl-2表达变化情况。结果:胃癌SGC-7901-SP细胞的增殖能力明显高于非侧群细胞(P<0.01);GS-Rh2能增强L-OHP对SGC-7901-SP细胞的敏感性,将细胞增殖阻滞在G1期(P>0.05),其Bax蛋白表达增高(P<0.01),Bcl-2蛋白表达降低(P<0.01),并与GS-Rh2浓度呈负相关(P<0.05)。结论:随着GS-Rh2浓度的逐渐增加,GS-Rh2逐渐提高了L-OHP对SGC-7901-SP作用的的敏感性,GS-Rh2可作为一种抗肿瘤药物的增敏剂。

       

      Abstract: Objective: To explore the effects of ginsenoside-Rh2(GS-Rh2) on the sensitivity of gastric cancer cells treated with oxaliplatin(L-OHP).Methods: The side population(SP) cells of human gastric cancers cell line SGC-7901 was screened using flow cytometry(FCM),the proliferation ability of SP cells was detected using CCK-8 kit.The half inhibition concentration(IC50) of GS-Rh2 and L-OHP on SGC-7901 SP cells were detected using MTT assay.The changes of cell cycle and apoptosis rate of SP cells intervened by GS-Rh2 combined with low concentration of L-OHP were detected using FCM,and the level changes of Bax and Bcl-2 protein in SP cells intervened by GS-Rh2 combined with L-OHP were measured using Western-blotting.Results: The proliferation capacity of SGC-7901 SP cells was significantly higher than that of non side population(P<0.01).GS-Rh2 could enhance the sensitivity of the L-OHP on SGC-7901-SP cells,block the cell cycle in G1 phase(P>0.05).The protein expressions of Bax and Bcl-2 increased and decreased,respectively(P<0.01),which was negatively correlated with the GS-Rh2 concentration(P<0.05).Conclusions: With the increasing of GS-Rh2 concentration,the GS-Rh2 gradually increases the sensitivity of L-OHP on SGC-7901 SP cells.GS-Rh2 can be acted as a sensitizer of anti-gastric cancer drug.
