    TRPV4对高血压小鼠胸主动脉内皮细胞钙离子稳态的调节作用

    Regulatory effect of TRPV4 on calciumion in thoracic aortic endothelial cells of hypertensive mice

    • 摘要: 目的:探讨瞬时受体电位香草素亚型4(transient receptor potential vanilloid type 4,TRPV4)在高血压小鼠胸主动脉内皮细胞中对于钙离子稳态的调节作用。方法:通过高盐饮食诱导小鼠高血压模型,分别在细胞和组织水平上使用钙离子荧光探针Flou-4/AM标记胸主动脉内皮细胞内钙,测定在TRPV4特异性激动剂GSK1016790A刺激下高盐饮食组和普通饮食组中细胞的钙离子浓度变化。结果:在胸主动脉内皮细胞的细胞和组织水平上通过使用GSK1016790A特异性激活TRPV4通道,普通饮食组和高盐饮食组细胞内钙离子浓度均增高(P<0.05),但和普通饮食组相比较,高盐饮食组在GSK1016790A刺激下钙离子荧光强度增幅低于普通饮食组(P<0.05)。结论:TRPV4参与了小鼠胸主动脉内皮细胞钙稳态的调节,通过激活TRPV4,促进内皮细胞外钙离子内流进入胞内,从而维持细胞内钙离子的稳态,而在病理模型,高盐饮食诱导的高血压状态下,TRPV4调节小鼠胸主动脉内皮细胞钙内流的作用受损。

       

      Abstract: Objective:To investigate the regulation of transient receptor potential vanilloid type 4(TRPV4) on calcium homeostasis in thoracic aortic endothelial cells of hypertensive mice.Methods:Intracellular calcium was labeled at the cellular and tissue level using calcium ion fluorescent probe Flou-4/AM,and the changes of calcium ion concentration of cells in high-salt diet group and normal diet group stimulated by the TRPV4-specific agonist GSK1016790A were measured.Results:The intracellular calcium concentration increased in the thoracic aortic endothelial cells of normal diet group and high-salt diet group stimulated by GSK1016790A to specifically activate the TRPV4 channel at cellular and tissue level(P<0.05).The increasing degree of the fluorescence intensity of calcium ion in high-salt diet group under the stimulation of GSK1016790A was lower than that in normal diet group(P<0.05).Conclusions:TRPV4 is involved in the regulation of calcium homeostasis in mouse thoracic aortic endothelial cells.Activating TRPV4 can promote the extracellular calcium influx into the cell to maintain the intracellular calcium homeostasis.In the pathological models,the effect of TRPV4 regulating intracellular calcium homeostasis is impaired in the hypertensive state induced by high-salt diet.
